Adjuvanthas an important role in the application of vaccines, with the development of modern molecularbiological technology, the research of adjuvant has been greatly promoted, for example, CpG ODNadjuvant,JSCOM adjuvantand TLR dependent adjuvant are eveloped.Athough adjuvants are variety, few adjuvants are approved for human, and the adjuvants which are globally recognised including aluminium adjuvant and MF59 adjuvant still have limitations. For example, aluminium adjuvant failed to stimulate an effectively cell immunity,what’ smore, aluminium adjuvant may cause local irritation response and adverse reaction.Therefore, it is significant to find safer and more effective adjuvants.This research is aim to investigate the effects of Low molecular weight heparin sodium (LMWH-Na) and pamidronic acid(PAD) as adjuvant on humoral immune response induced by hepatitis A virus (HAV) vaccine in mice.In this experimentlCR mice were randomly divided into different groups, and the mouse of each group are injected at 0 week respectively with live attenuated hepatitis A vaccine HepA-1 (18 EU) and different doses (100 μL,20 μL,10 μL,1 μL,0.1 μL) of LMWH-Na or (30μg,40μg,50μg,60μg) of PAD, while those injected with physiological saline as a negative control group (Negative control group), those injected with HepA-1(18 EU) as positive control group, those injected with Al (OH) 3(300 μg)+HepA-1(18 EU) as the aluminium adjuvant control group,200μl for each. The specific IgG level against HAV in sera of mice at weeks 4,8,12 and 16 after immunization were determined by indirect ELISA. During the experiment, the health status of the mice were observed, the kidney, spleen, liver, lung and heart of mice immunized with LMWH-Na 100 μLand PAD 60μg at the maximum dosage and those in negative control group were taken out after experiment, and prepared into sections for pathological observation. The results indicate that LMWH-Na can enhance the HAV antigen induced humoral immune response in mice,of which the levels increased as time went on, and reached peak values at week 8 then decreased gradually,but the titers of the anti-HAV IgG levels of various LMWH-Na groups isn’tdosage dependent,LMWH-Na lOμL is the most appropriate does. The anti-HAV IgG levels of various LMWH-Na groups were lower than the aluminum adjuvant control group.PAD have not enhance the HAV antigen induced humoral immune response obviously in mice, but this doesn’t mean the PAD do not have h the adjuvant effect,this result may be due to the young age of the mice,which results to the the immune systemimmature,and it may bealso due to the inappropriate choice of PAD dose,which results to fail to stimulate the effective immune response.In the next stage,we will investigate the possibility of PAD as adjuvant from this two aspects.In the animal safety testing,pathological sections showed the experiment is safe.The LMWH-Na 10μL group and the PAD 60μg groupwere not found abnormal,and the mice doesn’t appear tremble、seizures、convulsions and shock in the experiment.
