Chinese herbal medicine is a great treasure of Chinese.The heat-clearing and detoxifying drug is commonly used for antiviral, antibacterial, anti-inflammatory and regulating immune function. Along with the development of modern pharmacology, the research on the antibacterial and antivirus of Chinese herbal medicine has made great progress. Radix Isatidis、Honeysuckle、Mother chrysanthemum. Dandelion、Cypress、Chuan Huanglian、Selfheal、Forsythia and Elsholtzia bodinieri arr traditional Chinese herbal medicine in China which have a wide range of antibacterial, anti-inflammatory, antiviral, antioxidant and other biological activity, commonly used as clear pyrolysis poison. However, the research on the antibacterial and antiviral activity of these Chinese herbal medicines is not enough. In this study, antibacterial and anti HCV activity of the nine kinds of Chinese herbal of different polar parts were studied. And the anti HCV activity of Elsholtzia bodinieri was studied with active tracking method. Nineteen compounds were isolated from Elsholtzia bodinieri, and the anti HCV activity were evaluated of them. In addition, the antibacterial activity of the nine compounds which the large amount of compounds were evaluated too. The main research contents are listed as follows:In the study, we take nine kinds of traditional Chinese medicine as the test object. The active site was extracted by soaking in the ethanol and petroleum ether, chloroform, ethyl acetate, n-butanol extraction method. The antibacterial and anti HCV activity of the nine kinds of traditional Chinese medicine were studied. Hole method and the minimum inhibition concentration(MIC) experiment were used to test the inhibitory activity of five kinds of common pathogenic bacteria. The results showed that nine kinds of medicine in the coarse extract of gram positive bacteria (Staphylococcus aureus and Bacillus subtilis) inhibition effect is better than that of gram negative bacteria (Escherichia coli、Klebsiella pneumoniae、Salmonella enleritidis). And the effect of the antibacterial effect and the concentration of the dose effect relationship was obvious. Radix Isatidis, Honeysuckle, Mother chrysanthemum、 Dandelion、Selfheal、Forsythia and Elsholtzia bodinieri on live kinds of bacteria inhibitory activity part mainly concentrated in the petroleum ether extractive, chloroform extractive, ethyl acetate extractive and butanol extractive. There’s no antibacterial activity in the rafiinate. Cypress and Chuan Huanglian’ five extracts have antibacterial effect. On the basis of the establishment of HCV cell culture system, we take IFNα-2b and RBV as positive control which are the current standard treatment of HCV, to screen out anti HCV drugs in vitro using the method established in our laboratory. Nineteen compounds were isolated from Elsholtzia bodinieri, and the anti HCV activity were evaluated of them. In addition, the antimicrobial activity of the nine compounds which the large amount of compounds were evaluated too. The results showed that the chloroform extractive of Elsholtzia bodinieri, ethyl acetate extractive of Elsholtzia bodinieri, butyl alcohol extractive of Dandelion, butyl alcohol extractive of Cypress have better anti HCV activity.Elsholtzia bodinieri as the labiatae plants, which widely distributed in the southwest and northwest China. They usually use for prevention and treatment of a cold cough, headache, pharyngitis, such as fever and hepatitis. We adopt to activity tracking and fast separation method of combining the ethyl acetate extract of Elsholtzia bodinieri of anti HCV active compounds separation. A total of 19 compounds were isolated and they were evaluated for in vitro anti HCV activity. The large amount of 9 kinds of compound were studied by means of antibacterial activity. The results showed that there are 12 kinds of compound has certain activity of anti HCV (SI> 10). The compound 6 (Luteolin),9 (Rosmarinic acid),11 ((2R)-eriodictyol 7-O-β-D-glucopyranoside) (SI> 1000) have the best anti HCV activity effect, small cell toxicity and high HCV RNA replication inhibitory activities at low concentrations. Antibacterial experiments show that the 9 compounds have certain bacteriostasis effect, the strongest inhibitory effect on Bacillus subtilis, followed by Staphylococcus aureus. Compound 5 on 5 kinds of bacteria had the best antibacteria activity, the biggest bacteriostatic ring on Bacillus subtilis can reach 33.4 mm. MIC and MBC values were 0.781 mg/mL. The resistance of compound 5 to Salmonella enteritidis bacteriostatic ring diameter was 20 mm. It had antibacterial in a low concentration to 3.125 mg/mL. which the bacteriostatic ring diameter was 9 mm.
