Study On The Quality Standards Of Kadsura Coccinea

As a folk medicine, Kadsura coccinea belongs to the Kadsura genus, Schisandraceae family, whose main medical parts are its root and cane. In folk, it can be used for treatment of chronic gastritis, bruises, rheumatism and dymenorrhea because of its promoting blood and qi circulation and relieving swelling and pain. For years, researchers at home and abroad have focused analysis and corresponding activities on its chemical compositions.However, it is hard for K. coccinea to find a norm or standard still. Therefore, in this paper,a systematic and comprehensive research has been designed on the standard of K. coccinea for the first time.Firstly, 8 compounds isolated from the ethanol extracts of K. coccinea were identified with modern spectroscopic techniques. There are 7 lignans(kadsuralignan I 1,kadsuralignan J 2, schisantherin R 3, schisantherin S 4, binankadsurin A 5,Acetyl-binankadsurin A 7, dihydroguaiaretic acid 8) and 1 triterpene(kadsuralactone B 6),of which 3 and 4 were new compounds.Secondly, based on literatures, this paper chose binankadsurin A as a standard to compare the contents of binankadsurin A of K. coccinea produced in different places in China with HPLC method. Simultaneously, the reference sample was made by column chromatography on silica gel and DAC-preparation systems, with the purity of 96.06%identified by Q-NMR. In addition, according to 《Chinese Pharmmcopoeia》 published in2010, this paper also determined the content of extract, water, ash, heavy metal and pesticide residues of K. coccinea. The results show that: The content of biankadsurin A is between 0.74-6.93% because of K. coccinea produced in different palces, which should not be less than 0.7%; The content of water should be less than 10.0%; The content of ethonal extract should not be less than 12.0%; The total ash content should be lower than 5%,while the insoluble ash content should be less than 1.5%; The contents of lead, cadmium,arsenic, mercury and copper should not be greater than 5 mg/kg, 0.3 mg/kg, 2 mg/kg, 0.2mg/kg and 20 mg/kg, respectively; The contents of trichlorfon, dinethoate and omethoate should not be more than 0.1 mg/kg, 0.05 mg/kg and 0.02 mg/kg, respectively.In this paper, the relevant standands of K. coccinea were inquiried systematically and roundly for the first time, which laid the foundation of the use of K. coccinea safer and more effective, and the establishment of standards and related system of K. coccinea.
