Objective:This study was to detect microRNA-99a(miR-99a) and IGF-1R in renal cell carcinoma and renal carcinoma cell lines and to explore the role of the expression of microRNA-99 a and IGF-1R in renal cell carcinomaMethods:1Using Western blot, RT-PCR to detect the expression level of miR-99 a, IGF-1R in cancer cells and kidney tissue.2In vitro construction of miR-99 a mimics, IGF-1R-siRNA and NC-siRNA, and using the above-mentioned construction of plasmid to transfect cell lines 786-0.3Compare regulating miR-99 a expression and down-regulating IGF-1R expression in renal cancer cell proliferation, migration, and invasion and cell cycle by cell proliferation assay(CCK-8), detecting cell migration(scratch test), cell invasion detection(Trans-well) and flow cytometry.Redults:1.miR-99 a expression was significantly lower in renal cell carcinoma and renal cell carcinoma.2.IGF-1R expression was significantly higher in renal cell carcinoma and renal cell carcinoma. 3.Regulated miR-99 a and reduced IGF-1R could significantly inhibit the proliferation of renal cell carcinoma, survival and invasion and induce cell cycle G1 phase arrest.Conclusion:1.Our research shows that miR-99 a expression is significantly lower in renal cell carcinoma while IGF-1R expression is significantly higher and both of them participate in renal cell carcinoma and its development.2This suggests that miR-99 a and IGF-1R may function as a kidney cancer diagnosis and potential therapeutic targets.
