Objective:the aim of this study was to detect the expression level of collagen fiber and TGF-β1 in breast tissues and investigate the relationship between them,further explore the relationship between breast cancer ultrasound elastography (UE) elastic parameters with the content of collegan fiber and the expression level of TGF-β1,assessing breast cancer UE parameters reference value of disease progression and prognosis.Methods s Immunohistochemical method was used to detect 102 cases of breast cancer without receiving any treatment before surgery and 30 cases of adjacent normal breast tissue selected randomly,and then quantitatively accessed the expression levels of TGF-β1 in the breast lesions respectively. The specimen were stained with Masson trichrome method,collagen fiber area percentages were analaysed using a image processing software of plus 6.0. And then to investigate the relationship between TGF-β1 and collegan fiber, the clinicopathological characteristics of breast cancer such as histological grades,pTNM stage and lymph node metastasis.Strain elasticity examination and semiqualitative colour scores (1-5)were measured for each lesion before receving surgury. and further explore the correlation between the UE paremater and the content of collagen fiber, the expression levels of TGF-β1.Results:1. The collagen fiber was blue stained in the stromal of breast lesions by Masson method, and the collagen fiber area percentage in breast cancer tissues was significantly higher than in adjacent normal breast tissue (29.44+12.21 vs 20.32+8.28, P<0.01).2. the positive expression of TGF-β1 was brown stained in cytoplasm or cell membrane by IHC, and the expression level of TGF-β1 in breast cancer tissues was significantly higher than in adjacent normal breast tissue (P=0.000). TGF-β1 expression was positively correlated with histological grades,pTNM stage and lymph node metastasis (p=0.003,p=0.013,P=0.008),but no significant differences between TGF-β1 expression and age,tumor sizes (p=0.881,p=0.877). Positive correlation was found between collagen fiber area percentage and the positive expression of TGF-β1 protein in breast cancer tissues (r=0.747, p<0.01).3.102 cases of breast tumors, UE scoring:1 in 0 cases,2 in 7 cases,3 in 26 cases,4 in 43 cases,5 in 26 cases. the average content of collegan fiber were followed by 14.29±2.72,19.39±5.89,30.77±9.49,41.39±9.72 in different UE Scoring groups from 2 to 5,respectively.The content of collagen fiber within tumor with different UE scoring results was found to have a siginificantly difference. Further We found elasticity scoring of tumor is well correlated with collagen content of tumor (r=0.769,p<0.01)4.102 cases of breast tumors,the positive expression of TGF-β1:UE scored 2 in 4 cases,scored 3 in 9 cases,scored 4 in 36 cases,scored 5 in 24 cases.The data showed that high elasticity score (≥ 4) of breast cancers significantly increased the positive expression rate of TGF-β1 compared to those with elasticity score< 4 correlated with TGF-β1 positive expression (r=0.592, p<0.01).Conclusion:1. There were high expression level of TGF-β1 protein in breast cancer tissue, and positively correlates with collagen fiber.2. The TGF-β1 expression are also correlated with histological grades, lymph node metastasis and pTNM stage,suggesting that TGF-β1 might play important roles during the process of occurrence and evolvement of breast cancer.3. the UE scores is well correlated with the content of collagen fiber and the expression of TGF-β1,suggesting that the UE score level in a certain extent can reflect the expression of TGF-β1 protein,which the UE image may provide certain reference value for the biological characteristics and prognosis of breast cancer.
