Gelsmium elegans (loganiaceae) is an herbal medicine. It was firstly recorded in the book "Shen Nong’s Herbal Classic". The whole plant of Gelsmium elegans can be used for medicine with the effect of promoting blood circulation, subsiding a swellinrelieving pan, drawing out poison and killing insect. It is commonly used in the treatment of wet cold arthralgia, chronic osteomyelitis, internal hemorrhoids, ringworm and bruises embolism. Though it is resourcefully distributed in Southern China with Fujian Province as its main producing areas, the specific distribution is rarely reported. It has been reported in the literature that two to three kinds of main alkaloids of Gelsmium elegans had been determined by HPLC method, but 6 kinds of alkaloids in total were never measured altogether and so does the quality evaluation Therefore the current research can barely fully reflect the intrinsic quality of Gelsemium elegans.In order to clarify different origin and location> age of the quality difference,this paper specializes in conducting the herbal textual research and resource survey, studying the differences in medicinal ingredient among samples from different origins, different parts and different years. Construction of the system analysis method Gelsemium quality evaluation, analysis and comparison of the different sources of internal quality.1. Conducting a herbal research on the aspects of history, varieties, medicinal parts, collecting and processing, efficacy and clinical applications and so on in order to ascertain the variety, comprehend the resource distribution, folk or clinical application situation and so on, and to provide a theoretical basis for the following chapters of Gelsemium elegans Benth.2. On the basis of "the fourth National Census of traditional Chinese medicine resources", Conducting a resources survey on Gelsmium elegans in China, laying special emphasis on the distribution of resources in Fujian Province.Through field surveys to collect 37 pieces of wild Gelsemium samples of different origins of producing areas,127 pieces of samples from different parts, and different growth years up to 4 years, providing a reagent basis for alkaloids composition determination of Gelsmium elegans from 6 different origins in the later chapters.3. Utilizing Methanol as a base liquid extraction to analyze different extraction methods, different mutiple physic liquid extraction, different concentrations of methanol extraction, formic acid extraction contrast and different solvent methanol extraction contrast. Employing HPLC method to determine the 6 kinds of alkaloids composition, namely, content. Humantenidine、 Gelsemine、 Koumine、Gelsenicine、 Gelsevirine and Humantenine. Based on the results of experiments, best extraction method that can extract active ingredients of Gelsmium elegans with the highest extraction rate is selected, which is 125 time amount of liquid-to-solid ratio (0.2g crude drug,25ml extraction liquid),50% methanol (containing 0.1% formic acid), and ultrasonic extract for 30 minutes. This method provide for a relatively efficient extraction process of Gelsmium elegans samples in the later chapters.4、EsTab.lishing UPLC method to determine simultaneously 6 kinds of alkaloids composition contents in various medicinal parts (roots, the old stems, stems, leaves) of Gelsemium elegans of 14 batches from different producing areas.5、Employing UPLC-MS/MS method to determine simultaneously 6 kinds of alkaloids composition contents from all of the medicinal parts of Gelsemium elegans in different growth period (1-4 years). By experimenting, all of the studying and detection methods are in accordance with the analytic requirements. Determination results showed that:In addition to Humantenidine, another 5 kinds of alkaloids contents in different growth years, along withe total alkaloid, show a trend to increase in proportion to the number of their growth years.Tip Gelsemium alkaloids accumulate over time.It stems from the application of clinical and pharmaceutical industry,the old stalk maybe better than the yuang stalk of quality.
