Objective:Ischemic stroke is a kind of frequently occurring diseases with high morbidity and mortality, which is a serious threat to human health. With advanced technology of medical treatment means, the mortality of ischemic stroke has been reduced obviously, but the sequelae of this disease are still a great burden for those who after a near escape. A series of studies have recently shown that ischemic stroke caused by the damage nerve function is reversibl——the damaged tissues and cells are able to be regenerated. In this research, we use sprague dawley(SD) to make middle cerebral artery occlusion (MCAO) model as the experimental object. At the beginning, we will observe to what extent will the electro acupuncture adjust the ischemic stroke neurological symptoms. Then observe the effect of electro acupuncture to the ischemic brain tissues from a level of cell. Combined with the recently research progress in the field of regeneration of endogenous neural stem cell(eNSC), we are attempt to discuss the mechanism that how electro acupuncture treat ischemic stroke symptoms and promote the regeneration of nerve system. To provide the theoretical basis and clinical reference for the treatment of ischemic stroke with acupuncture.Methods:54 rats were randomly divided into electroacupuncture group, model group and sham operation group, each group was divided into 3 groups, each group was divided into 6 groups, each group was 3 days,7 days,14 days after the intervention. Electroacupuncture group and model group by zealonga method making rat right middle cerebral artery occlusion (MCAO) model, in the electroacupuncture group selected Baihui Point, Dazhui acupoint were given electro acupuncture treatment time 20 minutes, the model group not acupuncture, sham operation group only received surgical trauma. Respectively after 1 day, 3 days after modeling,7 days after modeling, modeling after 14 day on rat nerve damage symptoms score,7 days group rats were harvested after observation of the brain infarct size,14 day group rats were harvested after using immunofluorescence double staining on the right side of the ependymal membrane and the hippocampal dentate gyrus of nestin (the number of nestin and BrdU positive cells, and to evaluate the electroacupuncture for MCAO rat neural stem cell regeneration, and try to explore its mechanism. The experimental data were statistically analyzed by SPSS 20.Results:Compared with model group, in the electroacupuncture group in modeling for 7 days or less, the neurological impairment score no significant difference (P> 0.05); modeling 14 days later, the nerve damage symptoms scores with significant difference (P< 0.01).After 7 days of ischemia reperfusion injury, the infarct size in the electroacupuncture group (7D) was significantly smaller than that in the model group, and the difference was statistically significant (P<0.01).14 days after ischemia reperfusion injury, electroacupuncture group (14 rats in EA group, the dentate back and ventricular tube membrane of nestin and BrdU positive cells expression quantity is higher than the model group (14 days), the difference is statistically significant (P< 0.01).Conclusion:Electroacupuncture at "Baihui" and "Dazhui" can promote the recovery of neurological damage symptoms of MCAO rats and its effect on the brain ischemia reperfusion injury after 7-14 days when began to gradually obvious.Electroacupuncture at "Baihui" and "Dazhui" can reduce MCAO rat cerebral infarct area.Electroacupuncture at "Baihui" and "Dazhui" can promote the brain of MCAO rat hippocampal dentate gyrus, lateral ventricular membrane of endogenous neural stem cells proliferation.
