Background of the researchWith the improving of the incidence of metabolic syndrome and the changes of disease spectrum, The metabolic syndrome has become a major health problem in the world. Many studies have confirmed that metabolic syndrome was associated with cardiovascular diseases via multiple pathophysiology pathways. SIRT3 as nicotinamide adenine dinucleotide (NAD+) class Ⅲ histone acetylation enzyme off (histone deacetylases, HDACs) gene, participates in regulating mitochondrial energy metabolism, anti-oxidative stress, reduce the apoptosis, extend the life of the body and many other biological activities. At the same time, Researches have been confirmed that SIRT3 has a protective effect on myocardial in cardiovascular system. In previous study, we have found that the expression of SIRT3 in myocardial was closely related to left ventricular hypertrophy in hypertension mouses. The expression of SIRT3 significantly increased with the myocardial hypertrophy aggravated. However, few data was reported the relationship of between the SIRT3 and the metabolic syndrome with myocardial hypertrophy. Thus, we carried out the present study to explore the expression of SIRT3 in patients with MS and MS with myocardial hypertrophy, and to seek new therapeutic targets for the metabolic syndrome with myocardial hypertrophy.Purpose(1).To study the expression of SIRT3 in patients with metabolic syndrome.(2).To investigate the relationship between the SIRT3 and the components of metabolic syndrome.(3). To explore the relationship between the SIRT3 and MS with myocardial hypertrophy and its possible mechanism.Methods:1.Experimental patients and groupsWe collected 74 patients from the department of cardiology of the second hospital of shandong university. According to the 2007 China adult dyslipidemia prevention guide joint committee on metabolic syndrome diagnosis standards, and in 2007 the European hypertension guidelines of left ventricular hypertrophy diagnostic criteria (male LVM1> 125 g/m2, women LVMI> 110 g/m2),37 patients were taken as the group of MS. with an average age of 64.81 years,20 cases of men and women in 17cases; while the other 37 patients were taken as the group of MS with myocardial hypertrophy,with an average age of 65.95 years,20cases of men and women in 17 cases.31 cases of normal control group, They are healthy people, with an average age of 65.48 years,16 cases of men and women in 15 cases, Three groups matched in age and sex. by detailed history, physical examination, electrocardiogram, echocardiography and laboratory examination,nothing had been found., and exclusion of organic disease.2.Measurement criteria(1).A11 the following indexes, height, weight, blood pressure, blood sugar, TC, TG, LDL-C and HDL-C are measured in the next morning. The blood pressure was taken twice in the morning when the patient did not have any physical activity, and compute the average blood pressure.Height, waist circumference, hip circumference and weight, were measured.(2).Using the type of Ultrasonic echocardiography, Measuring diastolic interventricular septum thickness (IVS), diastolic end-stage left ventricular posterior wall thickness (PWT), left ventricular end diastolic diameter (LVD), left ventricular ejection ejection fraction (LVEF), and calculate the LVMI.3.Methods(1).Collecting the peripheral blood of patients and healthy, plasma and leukocytes was isolated.(2).Total mRNA was extracted from white blood cells, the mRNA reverse transcription into cDNA.(3).Application of real-time fluorescence quantitative PCR Technology, detecting the relative mRNA expression.(4)Detecting the plasma SIRT3 level by ELISA.4. StatisticsStatistical analysis was performed using SPSS 17.0 software, measurement data was expressed as means ± SD, measurement data was compared by the single-factor variance analysis and q-test. Comparison of two sets of measurement data with two independent samples T test. The enumeration data was compared by χ2 test. Linear correlation (Person-related) analysis studied the correlation between two variables. Values of p< 0.05 were considered statistically significant.Results1. To measure plasma SIRT3 by ELISA method, SIRT3was not detected. The result has showen that SIRT3 was located in cell.2. The expression of SIRT3 mRNA in the MS with myocardial hypertrophy,MS group increased significantly compared with control group. (P<0.01).3.The expression of SIRT3 mRNA in the MS with myocardial hypertrophy group increased significantly compared with MS group.4. The expression of SIRT3 was correlated negatively with TC、LDL-L、waist circumference(P<0.05). The expression of SIRT3 was correlated positively with LVM、LVMI(P<0.05), and negative correlation of LVEF(P<0.05).Conclusion1. Compared with the MS group, the control group, the level of SIRT3of MS with myocardial hypertrophy increased significantly.2. SIRT3 may be involved in the occurrence and development of metabolic syndrome and metabolic syndrome with cardiac hypertrophy disease.
