Deltamethrin belongs to synthetic pyrethroids. On the one hand, Deltamethrin has establishment of high efficient and structure stability, On the other hand,it has the pertinence effects on all effective object.Subchronic toxicity、 Acute toxicity、 Mutagenicity、 Teratogenicity was the toxicity of deltamethrin. There are some toxic symptom such as spasticity、salivation vomiting and limbs not adjusted.Recent studies to mechanism of deltamethrin concentrated on the electronic door of the sodium channel, but the specific target site is still disputed. We have found that the deltamethrin has good effects on C.elegans in the former work.As a result of C. elegans only have calcium channels. What is the mechanism of deltamethrin on C. elegans? whether or not is calcium channels?It was remain to prove that the role and the specific target site on c.elegans calcium channels of deltamethrin.We discussed the toxic symptom of C.elegans in deltamethrin of different concentrations.L4 stage ofC. elegans was exposed on 5、0.5 and 0.05 mg·L-1 deltamethrin in 24h,solvents and K-M solution for comparison. Respectively in the Asana microscope the offspring number、number of eggs、ovulation rate、body bending frequency and so on.As shown in the results of exposing 24h,Not only were reduced of the number of offspring and egg、ovulation rate in 30 mins,but also the head of bending frequency of the oscillation frequency and body significantly decreased compared with control group. It has close relationship with the concentration of deltamethrin according to the colonies rate dropped significantly of C.elegans and E.coli.The results of the difference of relative expression in of calcium channel α1 subunit egl-19,cca-1 unc-2 by the fluorescent quantitative PCR technology shown that the C.elegans was exposured in different concentrations can lead to gene expression change mentioned above three gen. It has biggest increase when the egl-19 exposed on the deltamethrin with 0.05 mg-L-1 for 12h,it reached to 12.5 times.While the cca-1 exposed on the deltamethrin with 50 mg·L-1 for 24h,it reached to 13.6 times.The unc-2 exposed on the deltamethrin with 0.05 mg·L-1 for 24h,it reached to 8.27 times.We figured out the role of deltamethrin on C.elegans site from the physiological and molecular level in this studied.Meantime, it provides a new basis for the developing new nematicides reagents.
