Objective:To investigate the toxic effects of paclitaxel?PTL?and dexamethasone?DEX?as single agents and two drugs in combination with Caenorhabditis elegans?C.elegans?to be exposed in its solutions.Firstly,to determine the median lethal concentration(LC50)of PTL,DEX,and the combination of two drugs in C.elegans by the acute toxicity test for 24h,then to explore the effects of PTL,DEX and the combination of the two drugs on the body length of the C.elegans with sub-lethal concentration,which was 1/10,1/100,1/1000 of 24h LC50.Methods:K-medium solution was used as a solvent to prepare PTL,DEX and two drug mixed solutions for experimental group.K-medium solution was the control group.Using liquid immersion method,the synchronized L4 C.elegans was exposed to the solution of the experimental and control groups for24 hours,the death and survival of the C.elegans were observed,while observing the growth,development,reproduction,and movement of theC.elegans.Probit regression model was used to calculate 24h LC500 and 95%confidence interval?CI?.In body length experiment,K-medium solution was used as a solvent to prepare different sub-lethal concentrations of PTL,DEX and a mixture of two drugs as the experimental group,K-medium solution as the control group.Using liquid immersion method,measuring the length change of the C.elegans after 60 hours of exposure.Results:?1?LC50of C.elegans was 374 mg·L-1(95%CI:369-378 mg·L-1)for 24h expose to PTL.?2?LC500 of C.elegans was 2471 mg·L-1(95%CI:24232515 mg·L-1)for 24h expose to DEX.?3?The LC500 of PTL was 226 mg·L-1(95%CI:207246 mg·L-1)and The LC500 of DEX was 1508 mg·L-1(95%CI:13811639 mg·L-1)when C.elegans were exposed to PTL and DEX mixed solutions for 24h.?4?After exposure to PTL at concentrations of 0.375 mg·L-1,3.750mg·L-1,and 37.500 mg·L-11 for 60 h.Compared with the controlgroup,the 0.375mg·L-11 exposure group had no significant difference in body length of C.elegans,it was[?0.809±0.047?mm vs?0.795±0.030?mm,?n=90,P>0.05?];the body length of C.elegans were inhibited in the 3.750 mg·L-11 and 37.500 mg·L-1exposed group,it were[?0.809±0.047?mm vs?0.489±0.027?mm/?0.276±0.022?mm,?n=90,P<0.01?].?5?After exposure to DEX at concentrations of 2.5mg·L-1,25.0mg·L-1,and250.0 mg·L-11 for 60 h.Compared with the control group,body length ofC.elegans were all inhibited in three groups.With increasing of exposure concentrations,C.elegansbodylengthwasshown the trend of shortening,it were[?0.809±0.047?mm vs?0.781±0.038?mm/?0.580±0.026?mm/?0.507±0.035?mm,?n=90,P<0.01?].?6?Afterexposure to PTL and DEX mixed solutions at concentrations of?1.50+0.23?mg·L-1,?15.00+2.30?mg·L-1,?150.00+23.00?mg·L-11 for 60h.Compared with the control group,body length of C.elegans were all inhibited inthree groups.With increasing of exposure concentration,C.elegans body length was shown the trend of shortening,it were[?0.806±0.021?mm vs?0.769±0.036?mm/?0.453±0.031?mm/?0.237±0.026?mm,?n=90,P<0.01?].?7?After exposure to PTL,DEX,PTL and DEX mixed solutions for60h,1.50 mg·L-11 and?1.50+0.23?mg·L-1,15.00 mg·L-1and?15.00+2.30?mg·L-1,150.00 mg·L-11 and?150.00+23.00?mg·L-11 pairwisecomparisonshave significant difference in C.elegans body length,it were[?0.787±0.020?mm vs?0.769±0.036?mm??0.702±0.037?mm vs?0.453±0.031?mm??0.553±0.035?mm vs?0.237±0.026?mm,?n=90,P<0.01?];0.23 mg·L-11 and?1.50+0.23?mg·L-1,2.30 mg·L-11 and?15.00+2.30?mg·L-1,23.00 mg·L-11 and?150.00+23.00?mg·L-1.The differences were[?0.786±0.020?mm vs?0.769±0.036?mm,?0.500±0.032?mm vs?0.453±0.031?mm,?0.331±0.025?mm vs?0.237±0.026?mm,?n=90,P<0.01?].Conclusion:PTL and DEX have acute lethal effects and developmental toxicity on C.elegans.The combination of the two drugs enhances the toxicity.PTL is more toxic to C.elegans than DEX.C.elegans can be used as an animal model of environmental toxicity monitoring of PTL and DEX.
