With the improvement of living standards, obesity and hyperlipidemia incidence increasing year by year, have become the main disease to harm to human’s health. Although common dugs of weight losing and lipid-lowering have certain curative effect, the long-term use will have side effect. Phytosterol esters(PSE) is edible safety. It can be dissolved in oil but not soluble in water and not easy to be dispersed to mixed with other substance. It can reduce blood lipid, the incidence of cardiovascular disease, etc. Kudzu is a traditional Chinese medicine, has been widely applied. Puerarin(Pur) is the extract of kudzu, powder and insoluble in water.It has reducing blood lipid, anti-inflammatory, anti-aging and other functions. Currently, rarely seen the reports of the combined use of phytostetol esters and puerarin about the effects of weight losing and lipid-lowering, so in the paper the nutrition obese mice model was established.The obese mices were divided into 5 groups(obese model group,phytosterol ester group, puerarin group,combined group and orlistat positive group). According to 10 times human dose,different groups were intragastriced for 6 weeks. By recording the weight,food intake, Lee index (Lee’s),fat coefficient and observing the slices of adipose and liver tissue to study the effects of weight losing of phytostetol esters and puerarin. Hyperlipidemic mice model was established and the mices were divided into 5 groups(hyperlipidemic model group, phytosterol ester group, puerarin group,combined group and simvastatin group).The positive group was lavaged with simvastatin according to 6 mg/kg-bw and the dose of other groups were in accordance with the dose of obesity experimental.Each group was intragastriced for 5 weeks. By testing serum lipid,atherosclerosis index,coefficient of liver and other indexs to study the effects of lipid-lowering of phytostetol esters and puerarin. Phytostetol esters and puerarin are difficult to dissolve in water. Phytostetol esters is not easy to disperse and long-term exposured to the air would be oxidized. Their value can be improved by the composite microcapsules of phytostetol esters and puerarin.The main results were as follows:(1)Phytostetol esters group, puerarin group and combination group could reduce the weight of obese mice, Lee index and fat coefficient. The weight of phytostetol esters group, puerarin group and combination group was significantly lower than model group(P<0.05). The combination group weight was significantly lower than phytostetol esters group and puerarin group(P<0.05), and compared with the positive group(orlistat group), there was no significant difference(P>0.05). The Lee’s of phytostetol esters group, puerarin group and combination group was significantly lower than model group(P<0.05). The combination group Lee’s was significantly lower than phytostetol esters group and puerarin group(P<0.05), and compared with the positive group, there was no significant difference(P>0.05). The fat coefficient of phytostetol esters group was lower than model group, but there was no significant difference(P>0.05). The fat coefficient of puerarin group and combined group was significantly lower than model group(P<10.05). Combined group was lower than phytostetol esters group and puerarin group, but there was no significant difference(P >0.05). The food intake of model group and the treatment group was no significant difference(P>0.05). The liver and fat slices showed that phytostetol esters group, puerarin group and combination group increased the number of fat cells within the same field of vision, recovered part of the structure of hepatic lobule, reduced fat vacuoles of liver cells, and the combination group was the most obvious effect. Results showed that phytostetol esters and puerarin had the effect of weight losing, and the combined use of the two had a reinforcing effect.(2)Phytostetol esters group, puerarin group and combination group had the effect of lipid-lowering. The serum triglyceride(TG) of phytostetol esters group, puerarin group and combination group was lower than model group, and there was a significantly difference between combination group and model group(P<0.05). Combined group TG was lower than phytostetol esters group and puerarin group, but there was no significant difference(P>0.05). The total cholesterol(TC) of phytostetol esters group, puerarin group and combination group was significantly lower than model group(P<0.05). combination group TC was significantly lower than puerarin group(P<0.05), but compared with phytostetol esters group and positive group(Simvastatin group), there was no significant difference(P>0.05). The serum high density lipoprotein cholesterol(HDL-C) of puerarin group and combination group was significantly higher than model group(P<0.05), and there was no significant difference between phytostetol esters group and model group(P>0.05). Combination group HLD-C was significantly higher than phytostetol esters group(P <0.05), and combination group was no significant difference with puerarin group and positive group(P>0.05). The low density lipoprotein cholesterol(LDL-C) of combine group was significantly lower than model group(P<0.05), The LDL-C of phytostetol esters group, puerarin group was lower than model group,but there was no significant difference (P>0.05). Combined group LDL-C was lower than phytostetol esters group and puerarin group, but there was no significant difference(P>0.05). Results showed that the combined use of phytostetol esters and puerarin had the effect of lipid-lowering and better therapeutic effect than alone.(3)The research results of composite microcapsule:the phytostetol esters and puerarin (mass ratio 2:3)were the core material. The best formula of composite microcapsule:the amount of 0.7%emulsifier, wall material ratio(Acacia Senegal:β-cyclodextrin) 5:5, core material/wall material 0.25, solids content 20%. The best conditions:inlet air temperature 180℃, outlet temperature 75℃, homogenization pressure 25MPa. Under the formula and conditions, phytostetol esters embedding rate was 89.04% and puerarin embedding rate was 80.15%. Composite microcapsule kept stable nature,good fluidity,moisture content 3.57%,density of 0.568g/ml,solubility of 95.11%.
