Objective:To investigate the effects of bone marrow mesenchymal stem cells(BMSCs) of emphysema in rats inflammatory cytokines in lung tissue and pathological changes, in order to investigate the effect of bone marrow mesenchymal stem cells of emphysema and the pathogenesis of emphysema.Methods: 3 SD rats were collected in the bone marrow cells suspension, the seeded in plastic culture flasks and cultured and inverted phase contrast microscope was used to observe the cell growth and morphological characteristics. 40 male SD rats were randomly divided into 5 groups: normal control group, emphysema model group, high concentration BMSC group, middle concentration BMSC group and low concentration BMSC group. In addition to the normal control group, the other 4 groups of rats were injected with PPE to induce the establishment of emphysema model, and the BMSC group was treated with different concentrations of BMSC in the tail vein injection. Rats were killed after 2 weeks of feeding, the pathological changes of lung tissue and alveolar lavage cells were observed, and the levels of IL-17 and IL-6 in BALF were detected by ELISA method.Results: 1. The emphysema model group and three groups of BMSC in BALF in total white cell count, the number of macrophages and neutrophil percentage were higher than in the normal control group(p< 0.05), three groups of BMSC group of low concentration of BMSC group percentage is higher than the number concentration in the BMSC group, the concentration in BMSC group is higher than that of the high concentration of BMSC group(p< 0.05). However, the total number of white blood cells, neutrophils and lymphocytes in the BALF of the 3 groups were lower than that of emphysema model group, the difference was statistically significant(p< 0.05).2. ELISA results showed that the three groups of BMSC group, emphysema model group compared with normal group, in BALF and serum levels of IL-17 and IL-6 increased significantly, with statistically significant differences(p< 0.05); 3 groups of BMSC group, BMSC group IL-17, IL-6 levels in low concentration was higher than that in concentration of BMSC group and in the concentration of BMSC group than high concentrations of BMSC group(p< 0.05). And the 3 groups of BMSC group and emphysema model group, IL-17, IL-6 levels were decreased, the difference was statistically significant(p<0.05).3. The BMSC group, pulmonary emphysema model group, the lung volume increased, emphysema like changes, BMSC group compared with the model group, the pulmonary emphysema changes significantly reduced and after the treatment of high concentration of BMSC group closest to normal state, good in concentration in the BMSC group, the concentration of BMSC group is better than that of the low concentration of BMSC group.4. BMSC group and the emphysema model group and normal group, increase the MLI, reduce the mans, differences were statistically significant(p< 0.05); three groups of BMSC group, low concentration of BMSC group than in the BMSC concentration group MLI increases, reduce the mans, increase the concentration of BMSC group higher concentrations of BMSC group MLI mans reduce(p< 0.05). The 3 groups of BMSC group and emphysema model group,MLI decreased, MAN increased, the difference was statistically significant(p<0.05).Conclusion: Intravenous injection of BMSC can improve the pathological changes of the rat model of emphysema, and the higher the concentration, the better effect, which may be through the regulation of the expression level of inflammatory factors to play a role.
