The Effect Mechanism Of Early Reversal Of Chemotherapy Induced POF In Rats By Serum Sex Hormone Level

Objective: drugs used in chemotherapy caused rat model of premature ovarian failure,through the analysis of the change of ovarian steroid hormone sythesis and physiological role(E2,P,t),ovarian reserve(FSH and LH),to further explore the left and Yougui bolus chmotherapy induced POF effect mchnim,reveal the scientific connotation of "the kidney controlling reproduction", provide effective clinical scheme for the establishment of TCM in the prevention and treatment of POF.Methods: 60,60 and 80 rats were randomly selected and included in the experi-ment,which were divided into three parts: early intervention,early late intervention and post intervention.The experiments were divided into six groups,the blank group,model group,Zuogui pill group,Yougui Pill group,about pill group and estrogen plus progesterone group.In the experiment,except the blank group,the other rats were intraperitoneal injection with cyclophos phamide,the initial loading dose of 50mg/kg,after the 8mg/(D/kg) dose of continuous intraperitoneal injection for 14 days.First part(early intvention)in the experiment,the blank group and model group with saline based,other groups use experimental drugs;to file in the estrous cycle as a behmark,estrus for medication,proestrus left Zuogui pill suspension and metestrus Yougui Pill mixed suspension,diestrus without medication,so periodic medication,a total of 15 days.The second part(early stage and late stage intevetion):methods and early intervention.Sixteenth days each group stopped the CTX,but continued to fill the experimental medicine for 20 days,for a total of 35 days.In the third part(late intervention),the CTX model was successfully used in rats,the experiment was made with early intervention, and the drug was used continuously for 20 days.The were the last treatment after 1-5 days will be in estrus or the 5 days had not yet appeared estrous rats to detect the serum sex hormone level,and through the homogeneity of variance test and single factor variance analysis.Results: 1. General: after modeling,the rats weight began to decline, and the blank group is statistically significant;after chemotherapy in different stages of experimental drug intervention;the body weight of rats have varying degrees of increase,the in between about traditional Chinese medicine gui pill sequential group and Western medicine of estrogen and progesterone group than single left gui pill group and single Yougui Pill group significantly;And the experimental longitudinal comparison to early combined intervention in the late most significant;Especially the general conditions(hair color,activi-ties,mental state,weight) change significantly.2.The rat estrous cycle:Western medicine estrogen plus progesterone group and traditional Chinese medicine about gui pill sequentialgroup, especially early intervntion in the late part of the, in improving the estrous cycle disorder POF model rats and restore normal cycle has significant effect.3.The rat serum sex hormone effects: in each group:with the blank group,model group rats serum sex horm-ones FSH,LH,E2,P is significant changes in experimental,statistical signif-icance;t have significant changes,indicating that the model was made succ-essfully. Through different stages of drug intervention,Zuogui pill group,Yougui Pill group, estrogen and progesterone group, about pill group the serum FSH,LH, E2,P,t have changed, which estrogen plus progesterone group, about pill group serum E2,P values increased significantly,FSH,LH were decreased significantly,t value significantly drop. About pill sequential group com-pared with Zuogui pill group, Yougui Pill group of steroidal hormones(E2,P level regulation has significant role; levels of FSH and LH have notably down regulated; single left gui pill group compared with Yougui Pill group have significant advantages in upregulated by E2 and single Yougui Pill group presentation left gui pill group have significant advantage in increasing the level of P. 2 the experimental longitudinal comparison: in early intervention and early synthetic late interventioncomparison, after making a successful model, through early synchronous intervention and early synthetic late inter-vention contrast, FSH,LH,E2,P,t have varying degrees of increase, especially in the early late intervention experiment, the western medicine group of estradiol and progestogen and about gui pill group sequential changes in the most significant. After modeling by late stage drug intervention and modeling early combination late intervention, rat serum sex hormones FSH,LH,E2,P,t have change,but early combination late intervention, serum sex hormone changes in rats is relatively more significant,more significant, especially in the early late intervention experiment, the western medicine group of estradiol and progestogen and about gui pill group sequential changes most significantly.Conclusion: 1 the POF model can be induced by cyclophosphamide in rats. 2 about the pill can effectively improve the rat’s emotional cycle.3 through the regulation of(E2,P,T) and the evaluation of(FSH, LH),can effectively regu-late the production of steroid hormones in ovarian granulosa cells and can improve the ovarian reserve capacity.
