The Influence Of XingNaoJieYu Capsule On PSD Model Rat’s Peripheral Blood T Lymphocyte Subsets

Objective:Establish post-stroke depression(post-stroke depression, PSD) model rat, to observe The influence of Xing Nao Jie Yu Capsule on PSD model rat`s peripheral blood T lymphocyte subsets. Method:1. Selecting 100 healthy Sprague Dawley(SD) rats, half male and half female, whose weight 200±20g.They were randomly divided into 8 groups: blank control group, blank group of Chinese medicine, blank group of western medicine, model group, early Chinese medicine intervention group, western medicine early intervention group, Chinese medicine treatment group, western medicine treatment group. 2. After a week of adaptive feeding, using the MCAO to design a permanent and focal Cerebral ischemia model,on the base of Cerebral ischemia model,combine the way to cure separatly and using Compound Reserpine Tablets to desigh the rat model of PSD except for blank control group, blank traditional Chinese medicine group and the control group of Western medicine. 3. Cerebral ischemia model was successfully prepared on the first day of the start given blank in Chinese medicine group, blank group of Western medicine, early Chinese medicine intervention group,western medicine early intervention group rats were intragastrically, 37 consecutive days;PSD the successful preparation of the model given Chinese medicine treatment group and Western medicine treatment group gavage for 16 days.4. The body weight and behavior of rats in each group were evaluated after 2 、15 and 37 days after modeling respectively. 5. At the end of the experiment, collecting the peripheral blood of rats to detect the contents of CD3+, CD4+and CD8+ by FCM and calculate the value of CD4+/CD8+. Result:1.The influnence of Xingnaojieyu Capsule on the weight and behavior of PSD rats.After desighing the model,compared with Blank control group, blank group of Chinese medicine and Western-medicine group, the weight, sugar consumption was obviously redused,so did the horizontal motion and vertical movement of rats.After treatment,compared with Model group,the indicators above were obviously higher,the most obvious was blank group of Chinese medicine early intervention group and Western-medicine early intervention group.The result showed Xingnaojieyu Capsule can make the weight,sugar consumption of PSD model rats increased,and improve the indicators of behavior. 2. The contents of CD3+, CD4+, CD8+and CD4+/CD8+ of peripheral blood T lymphocyte subsets in peripheral blood of rats in each group:(1)Compared with the blank group, the content of CD3+ and CD8+ in peripheral blood of model group was significantly lower than that of model group, and there was significant difference(P < 0.01).(2)Compared with the model group, the content of CD4+ in the early Chinese medicine intervention group, western medicine early intervention group, Chinese medicine group, western medicine treatment group increased obviously, the content of CD4+ in the early Chinese medicine intervention group and Western medicine early intervention group had no significant difference(P>0.05); the content of CD4+ in Chinese medicine treatment group and Western medicine treatment had no significant difference(P>0.05).(3)Compared with the model group, the content of CD8+ in the early Chinese medicine intervention group and the western medicine early intervention group increased, the early Chinese medicine intervention group have significant difference(P < 0.05) and Western medicine in the early intervention group had significant difference(P<0.01); there had no obvious difference in Chinese medicine treatment group,Western medicine treatment group and the model group about the content of CD8+.(4) Compared with the blank group, the peripheral blood CD4+/CD8+ of model group was significantly higher than that of model group(P<0.05). Conclusion:1.MCAO,curing separately,combine Compound Reserpine Tablets with building model can succeed in desighing the PSD model of rats.2.PSD model rats had some sympoms:the colour dark flatness,eat less,weight loss,lazy to move,slow activity,less sugur-consumption.Most of the sympoms which are improved after filled the Xingnaojieyu Capsule in the stomach,are same with the sympoms and pathological changes of PSD patients.3.Xingnaojieyu Capsule had no significant effect on rat model of peripheral blood CD3+ and CD4+/CD8+, it can increase the content of CD4+ and CD8 + on PSD model rats and the effect of early treatment on PSD rat is more significant.
