Objective This study objective aimed to investigate the effect and possible mechanism of tripterygium glycosides(TWP) intervene anti-angiogenesis drug sunitinib in culturing mice podocyte in vitro,to provide a theoretical basis for further animal experiments and clinical drugs using.Methods(1)Cultured mice podocyte in vitro,MTT assay was performed to examine the proliferation inhibition on mice podocyte of different dose of sunitinib for 24,48,72 h and sunitinib meanwhile treatment with TWP.(2)The apoptosis of podocyte in different groups after treated 48 h were observed by Annexin-V APC/7-AAD double-staining.(3) The expression of the associated proteins Nephrin,CD2 APin different groups were detected by Western blot.(4)The expression and distribution of Nephrin,CD2 AP were explored by immumofluorescence.Result(1)Being treated with various doses of sunitinib for 24,48,72 h,Sunitinib effectively inhibited the growth of mice podocyte in a doseand timedependent manner.(2) Compared sunitinib group,TWP+sunitinibgroupweresignificantly lower in proliferation inhibition rate after 24 h.However, there was no statistical siganificant difference in this two group after 24 h and 72 h.(3)A reduction in apoptosis rate was observed in sunitinib group compared with that of control group,and singnificantly increased in TWP+sunitinib group.(4)The Nephrin,CD2 AP expression of sunitinib group is lower than control group,and The TWP+sunitinib group increased while comparing with same dose of sunitinib group.
