Background and objective The Triptolide, which as a kind of Chinese herbal medicine, has a long history of good results on the treatment of various immunity diseases and a unique way of action. It is important in the Clinical Treatment because of the Multi-link immune suppression and anti-inflammatory. As a kind of basic immune depressants,TWP is applied in clinic generally including chronic glomerulonephritis,hydropigenous nephritis,nephritis of Schonlein-Henoch purpura,lupus nephritis,anti-exclusion of organ transplant and so on. It has a bright future in clinical application but it's adverse effects confine its wider clinicial application especially its disadvantage of genital system.。Estrogen- Progestogen sequential replacement therapy is often applied in the patient whose ovary was injured because of in critique age or operation and chems in clinic. Aimed at impairment of genital system induced TW,we used HPT in clinic and gained good consequence . To test our hypothesis, we design a suit of empirical method and expect to use this therapeutic regimen widespreadly in clinic and the study of drug.At the same time, some researchs indicate that the espression of bcl-2 and bax found on ovary granulosa cell(GCs) of the majority of living being ovary including the human being and rat participates of granulosa cell.The object of this study is to study the protective action of estrogen-progestogen replacement therapy (HPT) on female rats with impaired genital system induced by tripterygium wilfordii polyglycosidium (TWP) and its mechanism. Methods The 50 SD female rats were randomly divided into five groups: control group, intervention group, treatment group, recovery group and TWP group by body weight. The levels of serum gonadal hormone were detected by radioimmunoassay. The histological changes of the ovary were observed after HE staining. Using the immunohistochemical staining technique we detected the expression level of apoptosis regulatory protein Bcl-2 and Bax in ovarian granular cell. Results Compared with the control group, estrogen replacement therapy combined with progestogen could restrain these pathological changes produced by TWP, The level of E2 secreted by ovary was lower and the level of LH and FSH secreted by pituitary was higher in the TWP group and recovery group than the control group(P <0.05). And there was no significant difference between the control group and the intervention group (P > 0.05). To the effect of the Bcl-2 expressions in ovary cells of the rats, the TWP group was lower than the control group, and the intervention group and the treatment group were obviously higher than the TWP group, in the same time the expression of Bax protein showed the contrary change(P <0.01), among the total, there was no significant difference between the control group and the intervention group (P>0.05).Conclusion HPT can prevent ovary injury produced by TWP, whose mechanism may be at least partly correlated with up- regulation on decreased expression of Bcl-2 and suppression on increased expression of Bax...
