A Study On Reducing Inflammatory Response After Acute Ischemic Stroke By Ginkgo Ketone Ester Dispersible Tablets

Abstract/Summary:
[Objective] Observe the changes on expression levels of related inflammatory factors in serum after the treatment on acute ischemic stroke by Ginkgo ketone ester dispersible tablets, in order to evaluate its efficacy in anti-inflammation.[Methods] This study was a prospective, mult-icenter, clinical randomized and controlled trial. We randomly selected 52 patients with acute ischemic stroke and within 7 days of onset, whom were hospitalized in the department of neurology in Nanjing Drum Tower Hospital and Yangzhou First People’s Hospital during October 2012 to June 2014, and randomly divided them into therapeutic group (Ginkgo ketone ester dispersible tablets combined with aspirin group, n=27) and control group (aspirin group, n=25). Fresh blood was harvested before treatment (V0) and 12-14 days (VI) after treatment from enrolled patients, we detected concentration of inflammatory cytokines 1L-1β、IL-6、IL-15、IL-17A、IL-23 and TNF-a in serum with Milliplex Map multiple detection techniques, and carried on statistical analysis.[Results] 1. In 60 patients,8 cases (3 cases in therapeutic group,5 cases in control group) were excluded because of sample hemolysis, the shedding rate was 13.33%, so there were 52 cases (27 cases in therapeutic group and 25 cases in control group) left in all. There were no statistical differences in comparison of baseline datas such as gender, age, history of stroke, National Institute of Health Stroke Scale, blood pressure, liver and kidney function, four index signs of blood coagulation, blood glucose, blood lipid, and serum IL-1β, IL-6, IL-15, IL-17A, IL-23, TNF-a between the two groups of patients before treatment.2. The levels of serum inflammatory cytokines such as IL-1, IL-6, IL-15, IL-17A, IL-23 and TNF-a of all patients after treatment decreased compared with before treatment. Among them, the levels of IL-1β, IL-15, IL-17A, IL-23 and TNF-a of therapeutic group after treatment decreased significantly compared with that of before treatment (P<0.01), and the decline of IL-6 level also had statistical difference compared with before treatment (P<0.05); then the decline of IL-15 level in serum in control group after treatment had statistical difference compared with before treatment (P<0.05), but there were no statistical differences in the decline of other inflammatory cytokines levels compared with that of before treatment (P>0.05).3. Mean values of concentration differences of IL-1 β、IL-6、IL-15、IL-17A、IL-23、 TNF-a in serum before and after treatment in therapeutic group were larger than those in control group, it showed that decline ranges of inflammatory cytokines levels in serum after treatment in therapeutic group were larger than those in control group, and the mean values of concentration differences of IL-1p, IL-15, IL-17A and IL-23 in serum in therapeutic group before and after treatment had statistical differences compared with those in the control group (P<0.05), but there were no statistical differences in the comparison of concentration differences of IL-6 and TNF-a before and after treatment in two groups (P>0.05)[conclusions] The combination of Ginkgo ketone ester dispersible tablets and aspirin group can more reduce the levels of serum inflammatory cytokines compared with aspirin alone group in acute ischemic stroke patients, which instructs that Ginkgo ketone ester dispersible tablets can relieve the inflammatory injury by ischemic stroke, thereby it can better improve neurological functions of stroke patients.
