Objective: To study low-dose and subchronic iAs5+ exposure model, observe the effect of low-dose iAs5+ exposure to rabbits’ blood cells, blood lipid levels and the other related indicators of vascular endothelial and myocardial injury. Methods: 42 healthy male New Zealand rabbits were randomly divided into control group and 6 infected groups, the infected groups were given sodium arsenate solution, infected by free drinking water. Test weight weekly, after three months, kill animals after anesthesia, collected blood, urine and each viscera samples, weigh and record each visceras. To detect arsenic content of blood, urine, liver and heart by graphite furnace atomic spectrometry, with fully automatic blood cell analyzer to detect blood cells(WBC, RBC, PCT, et al),analyze liver function index(TBIL, DBIL and AST), blood lipid(TG, TC, HDL, et al) and myocardial enzymes(CK, LDH and HBDH) by automatic biochemical analyzer.ET-1, vWF sP-selectin et al were determined by ELISA kit. To test the expression of Nox2、Nox4 and NF-k B p65 by Western Blot. Results: The average of blood and urine arsenic in the infected group were higher than the control group, the differences were significant statistically(F=8.402,F=9.313,P<0.05). The WBC, LY, TBIL, IBIL and HBDH et al in the infected groups were higher than that in the control group, the differences between groups were significant statistically(F=10.921,F=3.161,F=8.258,F=6.971,H=13.898,P<0.05). Compare the infected group with the control group, the differences of ET-1、sP-selectin、TNF-α and IL-6 were significant statistically(F=70.225,H=31.015,H=35.255,F=14.033, P<0.05), ROS in serum of infected group higher than the control group(F = 3.881, P < 0.05), Nox2 expression of infected group in myocardial tissues is higher than the control group(H = 4.366, P > 0.05), NF- kB p65 expression is lower than the control group(F = 1.444, P > 0.05), but the differences were no significant. Conclusion: Expose to Low-dose iAs5+ can cause vascular endothelium injury and myocardial injury, there are also have influence on the expression of serum biochemical indicators and related proteins partly.
