Effects Of Sevoflurane Pre- And Post-conditioning On Hypoxia/Reoxgenation–induced Injury In Neonatal Rat Cardiomyocytes Cultured In High-glucose Culture Medium

Objective:To evaluate the effect of sevoflurane pre- and post-conditioning on hypoxia/reoxgenation – induced injury in neonatal rat cardiomyocytes cultured in high-glucose culture medium and the role of glycogen synthase kinase-3β. Methods:Primary cultured neonatal rat(1-3d after birth) cardiomyocytes were randomly divided into 12 groups:Normal groups use low-glucose(concentration, 5.56mmol/L) DMEM medium, hyperglycemia groups use high-glucose(concentration, 25mmol/L) DMEM medium. Control group(group NC/HC); hypoxia/reoxgenation group(group NH/R/HH/R); sevoflurane postconditioning group(group NS-Post/HS-Post); sevoflurane pre- and post-conditioning group(group NS-Pre+S-Post/HS-Pre+S-Post); SB216763 group(group NSB/HSB); Dimethyl sulfoxide group(group N-DMSO/H-DMSO). Cell survival rate, apoptotic rate, the LDH activity and the levels of phosphor-GSK-3β were measured at the end of the experiments. Results:In low glucose condition, sevoflurane pre- and post-conditioning group reduced the apoptotic rate, the LDH activity and increased the levels of phosphor-GSK-3β compared with NH/R group. However, in high glucose condition, sevoflurane pre- and post-conditioning group didn’t reduce the apoptotic rate, the LDH activity and the levels of phosphor-GSK-3β compared with NH/R group. Conclusion:Cardioprotection induced by sevoflurane is abolished by high glucose. GSK-3β may be involved with above processes.
