Objective: Through the observation of ethanol and dichloromethane extracts of Radix Piperis longi of experimental bile reflux gastritis(BRG) model in rat gastric mucosa pathological change, the level of serum gastrin(GAS)、TNF-α,gastric mucosal p21, PCNA, C-erbB-2 protein expression, and COX-2 mRNA expression level of gastric mucosa of different Piper longum organic solvent extracts on bile reflux gastritis of pharmacological effects and its mechanism. Methods: 80 SD rats were randomly assigned to normal group, model group, domperidone group, extract of ethanol, low, middle and high dose group,extract of dichloromethane, low and high dose group. Observe the pathological features of gastric mucosa, the level of serum GAS, TNF-αare measured by radioimmunoassay, The expression of P21, PCNA, C-erbB-2 in gastric mucosa is determined by immunohistochemical method, and COX-2 mRN A expression level of gastric mucosa by RT-PCR. Results: Compared with the model group, Radix Piperis longi extract reduced inflammation of gastric antrum tissue, increased the serum of gastrin and P21 protein expression, declined the serum of TNF-αand COX-2 mRNA expression level in different degree,the difference was statistically significant(P<0.05). Conclusion: Radix Piperis longi can improve bile reflux gastritis model rats gastric mucosa inflammation and may be related to the increase of serum GAS level and P21 protein expression, decline the serum of TNF-α and COX-2 mRNA expression level in gastric mucosa to further inhibit the progress of chronic inflammation.
