Objective:To investigate the association between HLA-DRB1 gene and the geneti c susceptibility and clinical features of breast cancer.To provide a clue to search fo r the susceptible/protective genes for breast carcinoma and explain the difference in its clinical characteristics.Methods:The case group was consisted of 198 breast ca ncer patients(98 Uygur and 100 Han patients),and the control group was consisted of 230 healthy people(115 Uygur and 115 Han women)diagnosed in the Affiliated Tumor Hospital of Xinjiang Medical University.Detailed clinical data were collected.PCR-SSP and CE were applied to determine HLA-DRB1 allele typing of all the sa mples.Stratification based on ethnic,Chi-square test was used to analysis the differen ces of HLA-DRB1 allele distribution frequency between the case and control group.The relationship between HLA-DRB1 allele and breast cancer’s clinical features wer e analyzed by Spearman correlation analysis.Results:The frequencies distribution of HLA-DRB1*01 and HLA-DRB1*16 were statistically significant in two groups(χ2=10.589,P=0.013,OR=3.377;χ2=12.408,P=0.006,OR=0.354).Stratified analysis:HLA-DRB1*01 had higher frequency in Uygur case group(χ2=10.180,P=0.026,OR=4.551),and H LA-DRB1*15 and HLA-DRB1*16 had lower frequency in Han case group(χ2=9.562,P=0.026,OR=0.481;χ2=9.463,P=0.039,OR=0.135).Allele subtype analysis:The frequenc y of HLA-DRB1*01:02 was significantly higher in Uygur cancer patients and the f requencies of HLA-DRB1*15:01 and HLA-DRB1*16:02 were significantly lower in Han cancer patients than that in controls(χ2=6.805,P=0.028,OR=3.038;χ2=9.535,P=0.006,OR=0.454;χ2=9.463,P=0.006,OR=0.135).HLA-DRB1 alleles which were associate d with the genetic susceptibility of breast cancer were no correlation with its clinic al features,such as age,pathological stage,immunohistochemistry,molecular typing and risk group classification and other factors(P>0.05 for all).Conclusions:HLA-DRB1*01:02 is the susceptible gene of Uygur patients,HLA-DRB1*15:01 and HLA-DRB1*16:02 are the protective gene of Han patients.HLA-DRB1 alleles which are associat ed with breast cancer genetic susceptibility are unrelated with its clinical features.
