The Protection Mechanism Research Of Jian Pi Shen Shi Prescription On Gouty Nephropathy

Objective: To observe the pathological intervention effect of jian pi shen shi p rescription on the renal function, renal index and kidney of model rat with go uty nephropathy from the perspective of treatment on patients with gouty nephr opathy with the use of Chinese herbal preparation, and observe the improveme nt of symptoms, to discuss the experimental and theoretical basis of the treatm ent on gouty nephropathy starting with spleen invigorating and dampness excret ing and eliminating phlegm and freeing channels.method: A gouty nephropathy rat model had been built, the rats were fed wi th high-yeast fodder and administered with adenine gavage, the rats administere d with high dosage of jian pi shen shi prescription + benzbromarone and the ones with moderate dosage of jian pi shen shi prescription + benzbromarone w ere taken as a treatment group, the rats administered with benzbromarone were taken as a positive control group, a blank group, model group, group of high and moderate dosage of jian pi shen shi prescription had been set as well, af ter 20 days of medication, the renal tissue specimen had been taken and admi nistered with tissue splice, HE staining, masson staining, immunohistochemical SABC staining, Quantify Real-time PCR, the level of blood uric acid, creatinine,serum urea nitrogen and renal index before and after experiment had been me asured, the effect of jian pi shen shi prescription on the level of blood uric ac id, creatinine, serum urea nitrogen and renal pathological change had been obse rved.Result: The group of high and moderate dosage of jian pi shen shi prescriptio n, high dosage of jian pi shen shi prescription + benzbromarone, moderate do sage of jian pi shen shi prescription + benzbromarone can equally significantly improve the blood uric acid, serum urea nitrogen, creatinine and 24-h urine m icroalbumin of the rat model of gouty nephropathy(p was equally less than 0.05), the group treated with jian pi shen shi prescription combined with benzbr omarone didn’t do a better job than the group treated with jian pi shen shi pr escription alone and the one with benzbromarone in reducing rat’s creatinine, s erum urea nitrogen and 24-h urine microalbumin; the group treated with jian p i shen shi prescription combined with benzbromarone did a clearly better job t han the group treated with jian pi shen shi prescription alone and the one with benzbromarone in reducing the rat’s urine acid, the group treated with jian pi shen shi prescription can down-regulate the expression of COX-2、TNF-α、TG B-β1 in the kidney of rats, and reduce the area percentage of renal interstitial fibrosis.Conclusion:Jian pi shen shi prescription can significantly reduce the blood uric acid, serum urea nitrogen, creatinine and 24-h urine microalbumin of the rat model of gouty nephropathy, and down-regulate the expression of COX-2、TNF-α、TGB-β1 in the kidney of rats, having a certain protective effect against the renal functional injury.
