1 ObjectiveShiqi exogenous compound consists of Gangmei, Tiebaojin, Putao, Choushimoli and Ludoule, which has the effects of dispelling wind clearing heat, relieving summer-heat and promoting digestion, used in the exogenous fever, cough, headache, ostealgia, dryness of throat and tongue, indigestion and its prevention of influenza effect is good. The safety and effectiveness of the prescription has already received a certain number of validation because of the prescription in Lingnan area has been used to prevent common cold and influenza nearly hundred years of history, but its pharmacodynamic material base is still unclear. In order to give shiqi exogenous compound more scientific and comprehensive theoretical foundation for clinical used and make full use of and dug up the compound potential medicinal value, this study was carried out on the shiqi exogenous compound more comprehensive efficacy confirmed and the pharmacological activity screening study of extracts from different technology.2 Methods2.1 The pharmacodynamics comparative study of shiqi exogenous compound extracts from different technology2.1.1 The preparation of extracts from different technology of shiqi exogenous compound:Use different technology to extract shiqi exogenous compound and concentrate the extracting solution to a thick paste standby in pharmacological experiment. The samples’numbers of the experiment are sample 1(shiqi exogenous particles), sample 2(compound extraction) and sample 3 (compound extration).2.1.2 Antipyretic experiment:Screen the best antifebrile extracts by the modle of endotoxin to New Zealand rabbit fever.2.1.3 Anti-inflammatory experiment:Screen the best anti-inflammatory extracts by xylene induced ear swelling model (Acute inflammation model)and the cotton induced granuloma (Chronic inflammation model).2.1.4 Analgesic experiment:Screen the best antalgic extracts by the hot plate experiment and the acetic acid-induced writhing experiment in mice.2.1.5 Antimicrobial experiment:detect different extracts’antibacterial activity and its minimum inhibition concentration(MIC) by Staphylococcus aureus.2.2 The anti-respiratory virus experiment in vivo and in vitro of shiqi exogenous tea extracts from different technology2.2.1 Anti-respiratory virus experiment in vitro:Observe the samples’ inhibiting effect on a variety of respiratory viruses on cytopathic effect induced by viruses by inoculating influenza virus A, respiratory syncytial virus, adenovirus, rhinovirus and parainfluenza virus into their sensitive cells respectively.2.2.2 Anti-respiratory virus experiment in vivo:Observe the samples’effect of infection to the lung index and mortality of mice caused by influenza virus A by the method of nasal drip.3 Results3.1 Antipyretic experiment:After given endotoxin the rabbits’model control group temperature significantly increase compared with the solven control group(P<0.01). The increase of the rabbits’body temperature of acetaminophen group in 1 h,2 h after the treatment is significantly lower than that of model control group (P<0.05). Compared with the model control group the samples groups of the group of the rabbit in the at different determination points after the treatment are no statistical difference of temperature rise in value (P>0.05)3.2 Anti-inflammatory experiment:The ear-swelling inhibition rate inhibition rate of sample 1, sample 2 and sample 3 are 20.07%,17.75%,15.97%; Compared with the solven control group, the cotton ball granuloma of sample 3 has significant difference (P<0.05) and the inhibition rate is 23.56%.3.3 Analgesic experiment:The hot plate pain threshold and body torsion times of the samples are no statistical difference compared with solvent control group.3.4 Antimicrobial experiment:Samples have no obvious inhibitory effect on Staphylococcus aureus.3.5 Anti-respiratory virus experiment in vitro:Samples under the non-toxic concentration are no effects on influenza virus A and rhinovirus; sample 3 under half the toxic concentration of 4 mg/mL can completely inhibit parainfluenza virus, under the non-toxic concentration of 2 mg/mL has 50% inhibitory effect on parainfluenza virus; sample 1 under 10 mg/mL can completely inhibit virus cause cytopathic effect, sample 3 under the non-toxic concentration of 0.5 mg/mL can protect 90% of the cells do not produce the cell lesion caused by virus. Sample 2 under the non-toxic concentration of 0.1 mg/mL and smaple 3 under the non-toxic concentration of 2.5 mg/mL can inhibit respiratory syncytial virus.3.6 Anti-respiratory virus experiment in vivo:The medium dose of the sample 2(0.42 g/kg) and the low dose of the sample 3(3.14 g/kg) can reduce the mice lung index, alleviate pulmonary consolidation, improve the symptoms that caused by viral pneumoni, and the inhibition rates of lung index are 15.19%, 15.82%; the high dose of the sample 1 (36.4 g/kg), the medium dose of the sample 2(0.42 g/kg) and the high dose of the sample 3(12.56 g/kg) can extend the survival time of mice and the life extension rate are 73.43%,87.30% and 72.43%.4 ConclusionTest study indicates that sample 1, sample 2 and sample 3 have certain inhibitory effect on acute inflammation caused by xylene in mice, sample 3 has certain inhibitory effect on chronic inflammation induced by cotton ball in rat.Sample 3 has certain inhibitory effect on adenovirus, syncytial virus and parainfluenza virus, sample 1 under the non-toxic concentration can inhibit adenovirus, sample 2 under the non-toxic concentration can inhibit syncytial virus.The medium dose of the sample 2 and the low dose of the sample 3 can significantly reduce the lung index of mice and has good inhibiting effects of influenza virus A in vivo; the high dose of the sample 1, the medium dose of the sample 2 and the high dose of the sample 3 can significantly extend the survival time of mice.
