Pharmacodynamic Study On Antiviral Activity Of Zedoar Tumeric Oil Eyegel Against Viral Keratitis

Zedoary Turmeric Oil (ZTO), a volatile oil, was extracted from the dryrhizome of Curcuma phacocaulis Valeton, Curcuma kwangsiensis orCurcuma phaeocaulis Valeton. Studies on Zedoary Turmeric Oil indicatedthat it had powerful therapeutic effects on many diseases of ophthalmology.At present, Zedoary Turmeric Oil gutta is widely used as a dosage form.Lots of clinical applications made sure that Zedoray Turmeric Oil gutta hadthe strong therapeutic effects on herpes simplex keratitis especially. Even forclinical application Zedoary Turmeric Oil gutta was prepared for one treat, notpreserved. Although the compounding was written in pharmacopeia, someunknown reasons of preparation process could affect quality and action of ZedoaryTurmeric Oil gutta. And the gutta was aqueous phase, it could not effect withfocus for infection, have not durg action. In this study experimenters comparedwith eye gel and gutta in terms of antiviral activities in vivo and in vitro tosupply experimental evidences for dosage form change of ZedorayTurmeric Oil.1 .Comparison of antiviral activities in vitro1) Evaluation of cytotoxicitiesThe cytotoxicities of experimental drugs against Hela cell lines wereexamined by the method of quantitative colorimetric MTT. It was shownthat the TC50 of eye gel form and gutta against two cell lines were bothhigher than 1000mg·L-1. The experimental data explained that of ZedoaryTumeric Oil eye gel had little drug toxicities.2) Comparison of pharmacodynamicsThe results of experiment indicated that the experimental druginhibited pathological changes in two cell lines which infected with HsvI,HsvII,Ad3, and intensity of the contribution was direct ratio to drug dose.In statistics analysis, the conclusion showed that logarithmic dose waspositively associated with protect rates for all drugs, Zedoar Tumeric Oileye gel showed a significant dose-effect dependence relation. Comparedtherapeutic index of experimental drugs against four viruses provided, eyegel carried out better antiviral effects than injection.2 .Comparison of antiviral activities in vivo1) Comparison of peroral dosage form and injection on curing viralpneumonia infected with HsvI,HsvII,Ad3.Compared effects of different experimental drugs on treating eyes ofrabbit, Zedoar Tumeric Oil eye gel was greatest among these groups andinhibited multiplication of HsvI,HsvII,Ad3, compared to viral controlgroup, effects of eye gel were best among all those of experimental druggroups also.In a word, on the point of pharmacodynamics, Zedoary Tumeric Oileye gel can be used as a new clinical dosage form of ophthalmology.
