Objective: In the study of Shabai Moistening Lung particles intervening in radiation-induced pulmonary fibrosis in rats,we observed pathological section and detected the level of TGF- β 1 in pulmonary tissue,Hyp in pulmonary tissue and plasma,LN and HA in plasma. The study will provide a new drug and provide a new idea of research for clinic against radioactive lung fibrosis.Methods: In the methord of irradiating on 2/3 zone of the right lung with 20 Gy 6MV x-ray all at once to make the rat model of the radiation-induced lung injury,setting-up blank control group with no irradiation or intervention; model group only by irradiating;dexamethasone positive control group given gavage with dexamethasone suspension for 4 weeks after molding;amifostine positive control group given single intraperitoneal injection of amifostine within 30 minutes before irradiation;three dose groups given gavage with low, medium and high level Shabai Moistening Lung particles solution for 12 weeks after molding.Six rats in each group would be killed in 2,12,26 weeks after molding,and we taked the right middle lobe for HE staining and Masson staining to observe pathological changes in radiation-induced lung injury clear., while analyzed the expression of transforming growth factor-β1( TGF-β1) in rat lung tissue ofradiation-induced lung injury by Reverse Transcription-Polymerase Chain Reaction(RT-PCR),the expression of hydroxyproline(HyP)in pulmonary tissue and plasma,laminin(LN)and hyaluronic acid(HA)in plasma by enzyme linked immune sorbent assay(ELISA).Results: 1. HE staining and Masson staining results of the lung tissue in each group:Blank control group showed no alveolitis or fibrosis.Model group showed mild- moderate alveolitis and mild fibrosis in the second week. Inflammatory cells infiltrated and widened alveolar septa, damaged alveolar structural, collagen fibers slightly increased in bundles. In the 12 th week, it showed severe diffuse alveolitis, there were diffuse infiltration of a large number of inflammatory cell in alveolar septa, alveolar structure collapsed in and merged together, collagen fibers moderately increased in banding. To the 26 th week, the range of the inflammatory lesions narrowed, alveolar structure continued collapsing in and merging together. These changes of high dose group were similar to amifostine group.High dose group of Shabai Moistening Lung particles didn’t show obvious inflammation and fibrosis in the second week; Mild inflammation and fibrosis appeared in the 12 th week; Inflammation lightened a little in the 26 th week and fibrosis didn’t become serious any more. These changes of high dose group were similar to amifostine group. Medium dose group’s degree of inflammation was similar to high dose group at each time point, and the degree of fibrosis showed moderate in the 26 th week. Low dose group showed mild alveolitis and fibrosis in the second week. In the 12 th week, it showed moderate alveolitis and fibrosis, the performance of the 26 th week was moderate alveolar inflammation and severe fibrosis. These changes of low dose group were similar to dexamethasone group.2.The expression of TGF-β1 in rat lung tissue analyzed by RT-PCR:The difference between high dose group and amifostine group had no statisticalsignificance(P >0.05). Compared with any other no blank group,they had the lowest TGFβ1 expression levels and significant difference(P <0.05).3.The expression of hydroxyproline in pulmonary tissue and plasma, laminin and hyaluronic acid in plasma analyzed by ELISA: The difference between high dose group and amifostine group had no statistical significance in pulmonary tissue hydroxyproline(P >0.05). High dose group had a lower expression level than amifostine group in hydroxyproline, laminin and hyaluronic acid in plasma.Compared with any other no blank group,high dose group and amifostine group had the lowest levels and significant difference(P <0.05).Conclusion: Shabai Moistening Lung particles can probably reduce the expression of TGF- β 1 and the generation of HyP 、 LN and HA, reduce the degree of radiation-induced pulmonary fibrosis.
