Objective: This article aims to investigate the expression level of microRNA-21 in renal cell carcinoma and its adjacent normal renal tissue with Real-time PCR, Combining with the clinical relevance,and to analyze its sense of expression levels and discuss the clinical significance for a farther study on its function and action mechanism.Methods:From 2015.08 to 2016.01, A total of 39 operation cases on renal cell carcinoma patients’ pathological diagnosed renal cell carcinoma(RCC) tissues and control tissues with adjacent nomal tissues(more than 2cm from the edge of tumor) were collected in tumor hospital affiliated of zhengzhou university(henan cancer hospital). The pathology of clinical,the TNM stage method were divided into four groups,includingⅠlevel andⅡlevel(n=18),Ⅲ level and Ⅳ level(n=21). In these examples,aged from 28 to 80 years(average 57.10±10.56 years), included 28 cases of renal clear cell carcinoma, and 11 cases 0f other types of renal cell carcinoma,24 males and 15 females.All patients were without any treatment. MicroRNAs extracted from 39 operation cases on renal carcinoma patients’ carcinoma tissue and its adjacent t normal tissue,will be detected preliminary by electrophoresis after RT-PCR to prove whether corresponding experiment conditions and primers designed are both in accordance with experimental requirements and then the expression of microRNA-21 was detected in the RCC tissue and adjacent normal tissue by quantitative real-time PCR. Finally,the relationship was also analysed between microRNAs and the clinical pathologic characteristics of patients with renal cell carcinoma.The clinical data were collected and analyzed by SPSS13.0 statistical software. P<0.05 indicated significant difference.Results: The result of the electrophoresis shows that miR-21 forms a single band,and its position and expressions in normal tissues and in the tissues of cancer are coincident with previous prediction.Compared with the control tissues,miR-21 has high expression levels in the tissues of renal cell carcinoma(39 cases), and the difference has statistical meaning(P=0.006﹤0.05). miR-21 has high expression levels in the tissues of clear renal cell carcinoma(28 cases),and the difference has statistical meaning(P=0.022﹤0.05). The expression levels of micR-21 in 21 of the 28 samples of renal clear cell carcinoma are obviously higher than its expression Levels in adjacent normal tissue and there are only 7 of the 28 simples where its expression levels are lower than its expression levels in adjacent normal tissue. there were apparent relationships between the expression level of miR-21 and pathologic types in renal cell carcinoma(P=0.031﹤0.05),moreover, the expression level of miR-21 between renal clear cell carcinoma group and renal chromophobe cell carcinoma group(P=0.010﹤0.05),renal clear cell carcinoma group and unclassified renal cell carcinoma group(P=0.040﹤0.05), the expression of miR-21 differences were statistically significant.However,express of miR-21 with the patient’s clinical staging, age and gender, Tumor location and size, smoking history,drinking history hadn’t statistical significance.Conclusions:The expression level of microRNA-21 in the tissues of renal cell carcinoma is obviously high,and the result suggests that microRNA-21 is relevant with the generation and the evaluation of renal cell carcinoma,it may have the function of oncogene in renal cell carcinoma,and it is expected to become a new target point for guiding the early diagnosis and the prognostice valuation of renal cell carcinoma.it also may be used as a potential therapeutic target in patients with RCC in future.
