Experiment Research Of Retention Enema With Original Prescriptions On The Influence Of Intestinal Mucosa’s Barrier System And Fecal PH Of Guinea Pigs

Objective: By copying bronchial asthma guinea pig model in this article,from PHysiology, pathology and behavior sides of guinea pigs from the quasi square four seasons clear out soup enema for young guinea pigs intestinal mucosal mechanical barrier and fecal PH effects,investigate the security of tra-ditional Chinese medicine enema,make traditional Chinese medicine enema application more scientific,standardized,for clinical services.Method: 50 healthy ordinary level guinea pigs,the experimental animals were randomly divided into 5 groups according to random number table method,A:healthy blank control group, model group B,C:the dexamethasone injection group,D:the four seasons clear soup enema high-dose group and group E the enteroclysis with clear four seasons out of small dose.Using 1% egg albumen ultrasonic atomization inhalation trigger asthma,before the treatment of enema,atomization 5 consecutive days;group A and group B using saline enema according to the 3ml/time,group C using dexamethasone to intraperitoneal injection according to 1 mg/Kg per day,group D according to traditional Chinese medicine(TCM) 21g/Kg enema,liquid retention for 5 minutes;group E take the same method using TCM enema 4.6g/kg per day,continuous dosing 1 week to measure drug withdrawal.Supervise the general condition of every group during atomization,terminal atomization score for each animal behavior after signs,with Switzerland’s(gram) dyeing,observation of guinea pig intestinal mucosal tissue PHysiological,pathological changes,the priority of infl-ammatory cells,goblet cells and mast cells.Results:① Behavior experiment results:compared with group A,atomization inhalation induced wheezing onset observed in group B,C,D,E guinea pigs are irritable,autonomous deep breathing becomes faster,limb frequent scratching nose and mouth,nodding respiration and other positive signs;Performance:after drug treatment group B score greater than 2 points, characterized like cough and choked by water,signs such as deep breathing fast and difficult;Compared with group B,group C score between 1 to 2 points, present a difficulty in breathing,drinking water accidentally choking by cough;Group D guinea pig asthma score between 0.5 to 1 points,characterized by shortness of breath,abdominal breathing,accidentally visible nose incitement;Group E guinea pig asthma score 1 minute,characterized by difficulty breathing.② In terms of mucosal mor PHology and structure compared with group A,group B microscopically intestinal mucosal epithelial damage, loss of integrity,can be obviously inflammatory cells infiltration around, the decrease in the number of goblet cells and intestinal tissue surrounding hyperemia and edema,mucosal damage level 3,C,D,E light pathological changes of the three groups compared with group B,mucosal form relatively intact, mucosal damage is level2,group D mucosal obviously thinning in the C and E group,no obvious advantage in reducing inflammatory cells,prompt Chinese medicine enema in a certain extent,can improve the pathological manifestation of diseased tissue,but if the drug concentration control is not ideal,it will cause secondary damage.③In the aspects of fecal PH change: group B compared with group A,fecal PH increased;C,D,E group compared with group A,fecal PH lower to some extent.Group B did not give any processing,fecal PH higher than other groups,after drug treatment, group C,D,E’s PH were decreased,group D drop more,group A and group B(P>0.05),no statistical significance,group B compared with group C,D,E,PH increased(P<0.01),with significant difference,compared group D and C,and E group(P<0.01),has obvious stati-stical significance,E group compared with group C(P>0.05),no significant statistical difference.Conclusions: Four seasons clear out soup enema can pull down the body’s PH,but the therapeutic effect of low PH did not show a greater advantage,it can weaken the mucous membrane barrier function;Enema on intestinal inflammatory change can be achieved moderate repair function,but the quantity of intestinal body fluid,factors such as temperature,concentration,velocity and pressure of mucous membrane potential damage,prompt enema has a risk,so we should think twice before taking actions,select a scientific and reasonable treatment plan.
