Objective To observe the effects of miRNA-16 on the proliferation and expression of VEGF-A of human lung adenocarcinoma A549 cells.Methods Human lung adenocarcinoma A549 cells were cultured and divide into three groups: miRNA-16 mimics group, negative control group and blank control group. Human lung adenocarcinoma A549 cells were transfected with 50nmol/L miRNA-16 mimics or negative control with Lipofectamine 2000(the condition of blank control group were not different from other two groups except the transfection situation). 24 hours after transfection, the expression level of miRNA-16 was measured using qRT-PCR. Cell proliferation at different time was measured by methylthiazol tetrazolium(MTT) assays. The expression level of VEGF-A of transfected A549 cells was tested with enzyme-linked immunosorbent assay(ELISA).Results qRT-PCR:24 hours after transfection and culture, the expression level of miRNA-16 in A549 miRNA-16 mimics group cells was significantly up-regulated compared with the blank control group and negative control group(P<0.05), the expression of miRNA-16 mimics group was 467.59±35.47 times higher than blank control group. The difference between blank control group and negative control group was not statistically significant. MTT assay: The proliferation of A549 cells was obviously inhibited(P<0.05) after tranfected with miRNA-16. The OD value of A549 cells was 0.471 ± 0.028(miRNA-16 mimics group), 0.561 ± 0.020(negative control group), 0.567 ± 0.025(blank control group) in 24h; 0.543 ± 0.021, 0.678 ± 0.026 å'Œ0.688 ± 0.022 in 48h; 0.623 ± 0.016; 0.895 ± 0.019; 0.907 ± 0.018 in 72 h. The difference between blank control group and negative control group was not statistically significant. ELISA: The expression of VEGF-A in A549 cells tested withELISA was obviously decreased in miRNA-16 mimics group(55.56 ± 6.04 pg/m)conpared with negative control group(152.07 ± 6.41 pg/ml) and blank control group(146.43 ± 7.71 pg/ml)(P<0.05). The difference between blank control group and negative control group was not statistically significant.Conclusions The findings of the study indicate that miRNA-16 may inhibits the proliferation of human lung adenocarcinoma A549 cells cultured in vitro,and may also down-regulate the expression level of VEGF-A in A549 cells.
