[Objective] To research the influence of the hydroxyapatite(HAP) nanoparticles toprolifrence and apoptosis of the human lung carcinoma cell line A549 in vitro.[Methods]①To make coating on the surface of the Propylend PTEF B with nano-Hydroxyapaptite coating. Compared with the Propylend PTEF B, to observe theadherance and growth inhibition and morphorlogic chang of the lung carcinomor cell lineA549 planted on the coating made of nano-hydroxyapatite under Inverted microscope②To observe the morphorlogic chang of the lung carcinomor cell line A549 from the twomaterials under transmission electron microscopy(TEM).③Nano-hydroxyapatite isdissolved in the medium in the concentration 12.5ug/ml,25ug/ml,50ug/ml,100ug/ml,400ug/ml. The lung carcinomor cell line A549 is cultivated in the medium. Contrastedwith the medium without hydroxyapatite nanoparticles, to oberve the growth inhibationby MTT method in 24h,48h,72h. To analysis the inhibiting rule of the lung carcinomorcell line A549 with the concentration and time changes.[Results]①Compared with the contrasting group, the cells on the coating of thehydroxyapatite decreased under Inverted microscope.②Unde TEM the cells exhibitcharacteristic of apoptosis, consist of cell membrane shrinkage, nuclear condensation andfragmentation, formation of apoptosis body as well as.③To detect the inhibition byMTT, the lung cancer cells is inhibited in the all concentration medium. The differencebetween 200ug/ml and 400ug/ml is not obvious(P=0.360).The difference between theother concentration is obvious(P<0.005). Hydroxyapatite nanoparticles inhibit the lungcarcinomor cells in a dose-dependent maner.The difference between 24h 48h and 72h isnot obvious for every concentration. The inhibition is not related with the changes of thetime.[Conclusion]①Obviously hydroxyapatite nanoparticles can inhibit the adherence and proliferance of the lung carcinomor cell line A549 in the vitro.②Hydroxyapatitenanoparticles can make apoptosis of the lung cells.③Hydroxyapatite nanoparticles:inhibit the lung carcinomor cells in the vitro in a dose-dependent mane.in some degree ofconcentration.
