Objective:To study the proteome of inferior colliculus (IC),identify the region-typical proteins of IC,explore the role of IC in the auditory integration and processing, which might be the candidate cause of the Central Auditory Processing Disorders.Methods:The proteome of IC cochlear nucleus(CN)、Superior Oliver Complex (SOC)、 the telencephalon (Rest) are consist of cytoplasm and cytomembrane proteins.2-D DIGE was used to separate and identify cytoplasm proteins and iTRAQ was used to identify and quantify the cytomembrane proteins,those with higher abundance in IC than other three were considered as IC-Region typical proteins,which may lead to functional specializations.Gene Ontology (GO) is an international standardization of gene function classification system. It provides a set of dynamic updating controlled vocabulary to describe genes and gene products attributes in the organism. The Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis is a collection of manually drawn pathway maps representing our knowledge on the molecular interaction and reaction networks.The protein-protein interaction (PPI) analysis is based on String database, it can reveal the interaction of proteins. GO, KEGG and PPI were used to analysis IC-region typical proteins.Results:With the use of 2-D DIGE,4 IC-Region typicalcytoplasm proteins were identified. Totally,1937 cytomembrane proteins were detected, among which there are 53 IC-Region typical cytomembrane proteins.These proteins have high abundance in IC,which may lead to functional specializations of IC. The GO analysis shows that the.se proteins mainly take part in transportation and localization, which will have effects on the regulation of neurons development and information integrations.KEGG analysis shows that these region typical proteins mainly take part in Oxidative phosphorylation,Glutamatergic synapseand GABAergic synapse. Through protein-protein interaction analysis, we find two candidate proteins, including CaMK2D and SV2A, which play significant roles in maintaining the balance of conducting and releasing of excitatory and inhibitory transmitters.Conclusions:(1) The IC-region typical proteins mainly take part in Glutamatergic synapseand GABAergic synapse,the mechanism of maintaining the balance of excitatory and inhibitory transmitters may be the key factor of information integrations.(2) CaMK2D and SV2A have high abundance in IC, this means that they play significant roles in IC. On the base of existing research findings,these two proteins mainly take part in maintaining the balance of conducting and releasing of excitatory and inhibitory transmitters, conducting further research to verify the relationship with Central Auditory Processing is necessary.
