| Objective: To investigate the effects of abnormal savda munziq(ASMq) on pathological changes and ultrastructural changes of dominating organs cells(brain, heart, liver) in rat model of depression. Methods male rat model of Depression 60, randomly divided into five groups: ASMq high, medium and low dose group(ASMq dose of 6.0,3.0,1.5 g / kg), a positive control group( fluoxetine, 3.5g / kg), depression model groups, normal control group, each dose group in 12 rats, intragastric administration once per day for 21 d. The depression model group was replaced by normal saline instead of ASMq. after the last administration, the animals were sacrificed by cervical dislocation,remove dominating organs fixed, HE staining and pathological changes of the "dominating organ" s were observed by the optical microscope, lead uranium staining, making conventional electron microscope specimens, scanning electron microscopy "dominating organ" s ultrastructural changes observed. Results Compared with the model group to the abnormal savda munziq high dose group "dominating organ" cells repair level increased, and the pathological changes of the injured tissue were significantly reduced. abnormal savda munziq low, middle dose group no significant difference in the pathological changes of the injured tissue. depression model group dominating organs besides the brain tissue ultrastructure of neurons, the remaining cells have different degrees of cell edema, decreased matrix density, cell ultrastructure of deterioration and other damage, but After ASMq intervention, brain edema decreased glial cells, extracellular matrix density, sparse organ celles significant recovery, significantly reduced neuronal synapses brain edema, showing slight vacuolar degeneration; decreased myocardial edema, cardiac extracellular matrix density was significantly recovery, no focal myofibrillar filaments were dissolved, while the inner mitochondrial edema decreased significantly, no local abnormal contraction band; hepatocyte edema decreased matrix density was significantly recovered slightly visible intracellular vacuoles, liver visible intracellular lipid droplets decreases, the cell gap widened significantly lower liver, bile capillary unexpanded, between the liver cells no collagen fibers. Conclusion The abnormal savda munziq is a protective and repairing effect on the "dominating organ" s rat model of depression pathological change and ultrastructural changes. |
