Studies On The Mechanism Of Action Of Anti-thrombus Of Abnormal Savda Munziq Extract

Objective:To investigate the effects of Abnormal Savda Munziq Extract(ASMq) against thrombosis and platelet aggregation.Methods:Adenosine diphosphate and collagen were used to induce platelet aggregation in rabbits to monitor the maximum ratio of platelet aggregation.Rats were used to establish the model of artery-vein bypass thrombosis and thrombus to measure wet weight.Contents of blood plasma TXB₂ and 6-keto-PGF1a were measured by radioimmunoassay.The coagulation time was measured by slide method.Results:ASMq not only could inhibit the platelet aggregation induced by ADP and collagen,but also could reduce the maximum ratio of platelet aggregation in rabbits.ASMq(10μg/ml,20μg/ml,40μg/ml)were incubated respectively for15,30 and 60 minutes,and the results indicate that platelet aggregation rates induced by ADP were 43.9%,36.0%,27.3%and 33.5%,24.2%,14.9%and 22.6%,15.4%,6.4%respectively,which shows that ASMq were dose-dependent and time-dependent.Platelet aggregation ratios induced by collagen were 44.6%,37.6%,27.6%and 30.2%,13.8%,19.6%and 23.7%, 33.7%,21.6%respectively.When the incubating time was 15 minutes,the inhibition rate of platelet aggregation was dose-dependent,While with the increase of time,it was time-dependent.ASMq[5.94g(herb)/kg,11.88g(herb)/kg]could inhibit artery-vein bypass thrombosis and the inhibition rates were 55.26%and 69.59%,dose-dependence was demonstrated as well.ASMq[5.94g(herb)/kg]can likewise increase the content of blood plasma 6-keto-PGF1a,but can not inhibit the release of TXB₂.ASMq[5.94g (herb)/kg]can prolong the coagulation time slightly.Conclusion:Abnormal Savda Munziq Extract exerts remarkable effects against thrombosis,and the mechanism of action is possibly concerned with the inhibition of platelet aggregation,increasing the content of blood plasma 6-keto-PGF1a and also with prolonging the coagulation time.
