Objective: to establish an early evaluation system for the effect of MRI in the treatment of colorectal cancer. We also want to evaluate the diagnostic value of MRI in the evaluation of tumor angiogenesis and the inactive state tumor angiogenesis, and provide a simple and feasible method for the development of anti tumor drugs. MRI imaging evaluation system, quantitative index and MRI scanning method were established in the early, effective and simple method to evaluate the clinical effect of MR. The evaluation function of MRI can be used as a biomarker to evaluate tumor angiogenesis and reflect the closure of tumor angiogenesis after drug treatment, and the related semi quantitative and quantitative parameters were obtained by DWI-MRI and DCE-MRI technology. Methods: We construct subcutaneous transplantation colorectal cancer in nude mice, respectively with avastin with 5-FU, endostar and 5-FU, Kanglaite and Xiaoaifei for the treatment of colorectal cancer in nude mice; monitory the transplanted tumor micro microvessel density and VEGF and PCNA, parameters, and evaluate the therapeutic effect and MRI monitoring results: 1) The model of subcutaneous colon cancer xenograft in nude mice as the research object, the traditional drug Iverson application at home and abroad approved in the treatment of advanced colorectal cancer anti angiogenic drugs An Weiting, endu injection and clinical colorectal cancer treatment drug 5-Fluorouracil Injection, Chinese medicine Kanglaite injection and Uygur Medicine Xiaoaifei, DWI were detected before treatment with different drugs and after treatment, different periods of DCE-MRI and b value, combined with the pharmacokinetic model, obtain the reflection of the semi quantitative and quantitative parameters of tumor cell metabolism and microvascular permeability of the tumor(peak enhancement, signal enhancement rate, maximum signal enhancement rate, peak time, slope value of Ktrans, Ve, Vp, Kep and ADC10 b, ADC3 b value, ADClow value, ADChigh value, ADCperf value), observe the changes of the above parameters, clear node whether there are differences between the observed indicators MRI carcinoma transplanted subcutaneously in nude mice in different treatment group, different stages of early, to evaluate the efficacy of anti angiogenic drugs and cytotoxic drugs. 2) MRI was used to examine at different drug treatment after 2 d, 7 d, 14 d half of the tumor bearing nude mice were killed, 10% neutral formalin were used to fix, paraffin package embedded, sectioned and stained with hematoxylin and eosin and parallel the immunohistochemical staining, including reflect tumor micro vascular density of the gold standard MVD determination and intensity of VEGF expression. Results: MRI was able to respond to the effect of treatment in time.Through studying the model of subcutaneous transplantation colorectal cancer in nude mice, the DCE-MRI and DWI determined before and after treatment can be used to obtain the quantitative parameters of tumor microvessel permeability(such as, Ve, Vp, ADCperf, Ktrans, etc.), then evaluate drug efficacy and analyze the correlation between the drug effects. MRI technology can be used as imaging biomarkers for the evaluation of the efficacy of anti tumor drugs, to provide early, fast, convenient and noninvasive clinical evaluation method for clinical rational use of drugs, and promote the development of anti tumor drugs. Conclusion: functional MRI has potential clinical application value and can be used as imaging biomarkers for the evaluation of the efficacy of anti tumor drugs, to provide early, fast, convenient and noninvasive clinical evaluation method for clinical rational drug use, and promote the development of anti tumor drugs.
