Objectives:To observation the effect of erythropoietin in the nerve differentiation of dental pulp stem cells. And to investigate the mechanism initially。To observe the effect of erythropoietin on the neural differentiation of dental pulp stem cells, and to initially clarify the mechanisms.Methods:Firstly, dental pulp tissue from healthy adult third molar is washed and cut into pieces under the aseptic conditions. After enzyme digestion,single cell suspensions were cultivated. Choose the passage 3 cells to seed into 6cm dishes. There are four group: DMEM/F12, DMEM/F12+EPO,RPE and RPE+EPO. Different group gives different medium. Finally, observe cellular morphology, measure cell viability by MTT and examine the gene expressions of nerve specific genes like Hes1 by Real-Time PCR。Results:Dental pulp stem cells are changed to neuron-like cell especially and shows a multipolar shape.Morphologically, dental pulp stem cells were spindle-shaped. After differentiation, they became round-shaped, with processes, showing a morphology of neuron-like cells. MTT assay showed that cell viability in group RPE group was simple in group RPE+EPO. Real-time PCR results showed that compared with DMEM/F12 group, the expressions of Map2,Hes1,Atoh7 were higher in DMEM/F12,RPE and RPE+EPO group. And the expressions of these genes in RPE+EPO group is higher than RPE group.Conclusions:EPO could promote dental pulp stem cells differentiation into ganglion-like cell.
