| Objective:To study the effect of Nutmeg extract on neurons and microglial cells in rats with vascular dementia (VD).Methods:Neuroethology was used to detect the impact of the changing of spatial learning and memory; HE staining was used to observe the effect of nutmeg extract on the arrangement and morphological changes of hippocampal neurons; Using immunohistochemical staining (OX-6 is an activated microglia Characteristic antibody), observe the effect of nutmeg extract on microglia activity.Results:1.Morris water maze test results show that Learning ability and memory function of control group have obvious advantages than other groups, treatment group compared to the model group showed significant progress, escape latent period and the rate of passing through the target quadrant was significantly better than the model group, the spatial learning and memory have been significantly improved.2.HE staining showed that the control group hippocampal pyramidal cells in condition that uniform size, in alignment, uniform color, clear nucleolus; Model group hippocampal pyramidal cells arranged in disorder, the cell gap widens, signs of neuronal degeneration; The nutmeg extract treatment group neuronal morphology approximate normal group, significantly restore and improve.3. Immunohistochemical staining showed that the cerebral cortex and hippocampus region of the model group visible to a large number of OX-6 antibody stained brown granule; The positive cells of nutmeg extract treatment group were significantly reduced and in shallow staining.Conclusion:1. Nutmeg extract can effectively alleviate and improve the cognitive dysfunction and damage in rats with vascular dementia (VD).2. Nutmeg extract can inhibit the damage of hippocampal neurons induced by hypoxic-ischemic, has a neuroprotective effect.3. Nutmeg extract can inhibit the activation of hippocampus microglial cells in rat with vascular dementia, has anti-inflammatory effects. |
