Chemometrics is a branch of chemistry.The purpose of Chemometrics is to extract the maximum useful chemical information from a large number of chemical measurement data. Second-order calibration method in Chemometrics can relies qualitative and quantitative analysis of analytes even in the presence of unknown disturbances by using mathematics separation instead of physical separation or chemical separation. Many analytes can produce natural fluorescence. We detected the contents and recovery rates of active components in traditional Chinese medicine and human plasma by using three-dimensional fluorescence spectroscopy coupled with second-order calibration method.In Chapter 2, Three-dimensional fluorescence spectroscopy coupled with second-order calibration method based on alternating trilinear decomposition(ATLD)algorithm, detected the contents and recovery rates of Synephrine in Citrus. Using the new method, we obtained satisfactory qualitative and quantitative analysis even in the presence of an unknown disturbances in Citrus. The results obtained by high performance liquid chromatography(HPLC) were in good agreement with the results of the new method.In Chapter 3, a new method for determination of Mangiferin in Mango leaves and human plasma without pretreatment even in the presence of other uncalibrated interferents by using excitation-emission matrix fluorescence combined with second-order calibration method based on alternating trilinear decomposition(ATLD)algorithm. We got satisfactory results. The results obtained by high performance liquid chromatography(HPLC) were in good agreement with the results of the new method.In Chapter 4, the contents and recovery rates of Eugenol were detected in Clove by using three-dimensional fluorescence spectroscopy coupled with second-order calibration method based on alternating trilinear decomposition(ATLD) algorithm.Excitation spectra and emission spectra of Eugenol and unknown interferences in Clove were overlapped seriously, so we can’t detect Eugenol contents in Clove directly. Owing to the second-order advantage achieved by the proposed method, we obtained satisfactory qualitative and quantitative analysis even in the presence of an unknown disturbances in Clove. The results obtained by high performance liquid chromatography(HPLC) were in good agreement with the results of the new method.
