Experimental Study On Euthyroid Sick Syndrome Of Organophosphorus Pesticide Poisoning In Rats

Objective: In this study by measuring the omethoate poisoning rats serum thyroid hormone concentration, serum TNF alpha concentration, serum NF kappa B concentration, explore AOPP rats would happen euthyroid sick syndrome; whether thyroid function changes were correlated with the degree of poisoning、the degree of pulmonary edema and serum TNF alpha levels. In order to judge the prognosis of patients with AOPP and provide theoretical basis for the new target treatment of AOPP patients.Methods:84 healthy male SD rats were randomly divided into control group(Group C,12), low dose poisoning group(Group L,36) and high dose poisoning group(Group H,36). Group C gavage with sterile saline, Group L gavage with 1/2 the half lethal dose of omethoate(22.5mg/kg),Group H gavage with the half lethal dose omethoate(45mg/kg). Respectively after exposed 0.5h, 3h, 6h, 12 h, 24 h, 48 h killed the rats. Determination of thyroid hormones(TT3, FT3 TT4, FT4, TSH,RT3)in the serum of rats, serum TNF alpha concentration, serum NF kappa B concentration, lung coefficient, thyroid levels of TNF alpha in each group and observe the thyroid pathological changes of rats.Results: 1、Group L、Group H rats serum TT3, FT3 concentration decreased、r T3 concentration increased(P < 0.05), Group H rats serum TT3, FT3 concentration below Group L, r T3 concentration is higher than that of Group L(P < 0.05); Group H rats serum TT4, FT4 concentration was lower than that in Group C、Group L(P < 0.05), Group L rats serum TT4, FT4 and Group C compared to no significant difference(P > 0.05); three groups of rats serum TSH concentrations no significant difference(P > 0.05).2、Group L, Group H serum TNF alpha concentration, serum NF kappa B concentration, lung coefficient, thyroid levels of TNF alpha increased(P < 0.05),Group H were higher than Group L(P < 0.05). The serum TNF alpha concentration and thyroid levels of TNF alpha were positively correlated(r = 0.708, P < 0.01), the serum TT3 concentration and serum TNF alpha concentration were negatively correlated(r =-0.69, P < 0.01), the serum TT3 concentrations and lung coefficient were negatively correlated(r=-0.86, P < 0.01).3、Thyroid pathological changes: thyroid follicular of Group C were oval shaped distribution, the size of the distribution is roughly uniform, the follicular cavity in the pale red glial uniform distribution, a small blood vessels around the follicle at each time. After 0.5h, Group H start with edema around the follicle, telangiectasia, intravascular red cell aggregation, this reached to the peak at 12 h, after 24 h edema, capillary expansion eased, after 48 h were not returned to normal, the exposure to 0.5h to 48 h appear by gradually aggravated the thyroid follicular size, number of follicular cavity glial emptying, fused follicles, follicularepithelial cells arranged in disorder. Group L rats thyroid pathological changes were similar with the Group H, but the edema, the degree of capillary dilation were lighter.Conclusions: 1.Euthyroid sick syndrome appears in rats with acute organophosphorus pesticide poisoning. The change of serum thyroid hormones were related with the severity of poisoning.2 、 Serum TT3 concentration was negatively correlate with lung coefficient and serum TNF alpha concentration, serum TNF- alpha concentration was positively with the level of thyroid TNF- alpha。3、TNF- alpha 、NF-κB may be the important factors of euthyroid sick syndrome in rats with acute organophosphorus pesticide poisoning.
