Objective:This study aimed to explore the effect of acupuncture on CRS mice and its part of molecular biology mechanism by observing acupuncture intervention in nasal mucosa morphology in model mice of CRS and the influence on Eotaxin-1, iNOS expression. At the same time, To provide scientific experimental basis for clinical application of acupuncture and moxibustion for prevention and treatment of CRS.Methods:160 male C57BL mice were completely randomly divided into 8 groups.This paper only incorporates the datas of acupuncture group,model group,sham operation group,normal group,western medicine group.To build chronic sinusitis models in Acupuncture group, western medicine group, sham operation group,normal group, model group according to the surgical operation method in Streptococcus pneumoniae,normal group don’t received intervention,Bacteria were not placed in the sham operation group.Begin to treatment from the fifty-seventh day.In the western medicine group, the mice were treated with clarithromycin tablets 0.103g/kg.d,once a day. In the acupuncture group, the mice were treated with needling the bilateral Zusanli (ST36),neutral supplementation and draining, the needle for 20 minutes,once every other day;with mild moxibustion on Guanyuan (RN4) for 20 minutes,once every other day. The sham operation group, model group, normal group, distilled water,0.2ml/only-day,The treatment lasted for 14 days. To observe the morphological changes of nasal mucosa with HE staining and detect the expression of Eotaxin-1 and iNOS by immunohistochemical method, with the average integral optical density value of expression.Results:1. symptoms and physical signs:At the end of treatment, The sham group and the normal group had no nasal congestion, runny nose, sneezing, scratching nose and other symptoms; there is a certain degree of the symptoms of model mice, mice with partial tearing, conjunctival hyperemia and edema; acupuncture group and Western mice nasal congestion, runny nose, sneezing,scratching nose and other obvious symptoms, no epiphora and conjunctival hyperemia edema.2. The result of nasal mucosa morphology with HE staining:In normol group, Nose (sinus) mucosa epithelial cells arranged in neat rows,the sinus mucosa cilium epithelium structure of mice was normal. There was no inflammatory cell,no gland hyperplasia,a small amount of goblet cells scattered in the meantime.In model group: nose mucosa showed the changes of chronic inflammation,the epithelial cilia arranged in disorder,In the part of region,the cilia got flattened and became necrotic,The hyperplasia of fibrous tissue and glands distributed in the submucosal.The Lymphocyte and Plasma cell showed obvious Infiltration;Each group of chronic inflammation of the nasal mucosa arranged in the order from heavy to light:The model group>The western medicine group> The acupuncture group> The sham group. The high expression of Eotaxin-1 participated in the pathogenesis of CRS.3. ①The expression of Eotaxin-1:-Compared with the normal group,the expression of Eotaxin-1 of model group was increased (P=0.000), the difference was statistically.Shows that the hight expression participated in the pathogenesis of CRS. The average integral optical density value of Eotaxin-1 in acupuncture group lower than the model group (P=0.000), through reducing the expression of Eotaxin-1, acupuncture and moxibustion may play a important role in preventing the formation of nasal polyps.②The expression of iNOS:Compared with the normal group,the expression of iNOS in each group were increased (P<0.01),the difference was statistically significant,Shows that the hight expression of iNOS participated in the pathogenesis of CRS. Compared with the model group, the average optical density of iNOS in acupuncture group was significantly lower (P=0.000),compared with the sham operation group,it had no significant difference (P>0.05),Acupuncture and moxibustion could regulate the expression of iNOS in nasal mucosa, and suppress the chronic inflammatory injury of the nasal mucosa.Conclusion:1. Acupuncture could effectively ameliorate the symptoms of CRS mice, repair the damage of the nasal mucosa’ inflammatory injury on CRS. On the pathological morphology, the effect of acupuncture on the repair of nasal mucosa was better than western medicine group.2. In the process of the disease of CRS, the expression of Eotaxin-1 and iNOS in mouse nasal mucosa became greater, The therapeutic effects of acupuncture and moxibustion on CRS resulting in lowering the expression of iNOS and Eotaxin-1.At the same time, through reducing the expression of Eotaxin-1, acupuncture and moxibustion might play a important role in preventing the formation of nasal polyps.
