Objective: TO investigate the changes and the relationships of interleukin-32(IL-32),matrix metalloproteinase-13(MMP-13) in rheumatoid arthritis(RA).IL-32 and MMP-13 were detected by enzyme-linked immunosorbent assay(ELISA), the correlation analysis were done among above results and rheumatoid factor(RF), anti-CCP antibody, erythrocyte sedimentation rate(ESR), hypersensitive C-reactive protein(hs-CRP), disease activity score in28 joints(DAS28), duration and so on. This study revealed effects of IL-32 and MMP-13 on the pathogenesis of RA and provided applications for further study of RA and its clinical treatment.Methods: 136 patients with RA and healthy persons were selected from August 2014 to April 2015, all of these patients were consistent with the American College of Rheumatology(ACR)/European Union of resistance Rheumatism(EULAR)RA classification standards revised in 2009/2010.82 healthy persons were selected as the control groups, statistical analysis showed that between the two groups of age, sex matched. DAS28 was used to judge the RA disease activity, and X-joint damage stages and functional groups according to ACR criteria were divided into four. The serum of all the persons were collected and stored.The concentrations of IL-32, MMP-13 and anti-CCP antibodies were detected by ELISA, RF and hs-CRP were analyzed by immunoturbidimetric assays. Use t test, single-factor analysis of variance and LSD test o analyze the levels among the different groups, the correlations among different parameters were analysed by pearson.Results:1 Comparisons of serum IL-32 and MMP-13 between the RA and control groups,the levels of IL-32 and MMP-13 in RA group were(135.42±30.57)pg/ml,(16.78±3.14)pg/ml, while the control group were(117.35±29.76)pg/ml,(15.09±2.43)pg/ml, the two groups were statistically significant(P<0.001,P<0.001).2 Comparisons among the different active groups in RA, the levels of serum IL-32 and MMP-13 in RA active group were higher than those of stable group(P<0.001,P<0.01), mild and severe active groups were significantly different(P<0.001,P<0.01).No significant differences were found among the duration groups(p>0.05, p>0.05).3 Comparisons of serum IL-32 and MMP-13 among the different joint X-ray stages, there were differences between â…£ andâ… (P<0.01,P<0.001), â…¢andâ… stages(P<0.05,P<0.01), while the levels of MMP-13 in â…£ higher than that of â…¡ stage(P<0.05).4 The levels of IL-32 and MMP-13 among the functional groups were significantly different, especially between â…£ andâ… (P<0.01,P<0.001),â…£ andâ…¡(P<0.05,P<0.01), â…¢ and â… functional group(P<0.05,P<0.001).In â…¢ group,MMP-13 was higher than that of â…¡functional group(P<0.05).5 Correlation analysis between IL-32, MMP-13 and clinical data in RA group, IL-32 and MMP-13 had positive correlations with ESR, hs-CRP, RF,anti-CCP antibodies, DAS28(P<0.001 or P<0.01), and correlation of IL-32 and MMP-13 had statistically significance(r=0.592,P<0.001).Conclusions:1 The levels of serum IL-32 and MMP-13 increased in RA patients, they played an important role in the development and progress, and they had significant positive correlations with RF, anti-CCP antibody, ESR, hs-CRP,DAS28, the tender and swollen joint numbers of RA.2 With the stages of RA activity, X-ray and function increased, IL-32 and MMP-13 elevated, they may be as important markers to assess disease severity of RA. |
