The Effect Of BMSCs And HGF On Regeneration Of Hair Follicle

Aim:To study the effect of bone marrow mesenchymal stem cells and hepatocyte growth factor on regeneration of hair follicle.Methods:First, a kind of collagen/chitsan scaffold was fabricated. Then the collagen/chitsan scaffold being incorporated with BMSCs or HGF was treated as control groups respectively, and the collagen/chitsan scaffold with both BMSCs and HGF was treated as experiment group. After that, the collagen/chitsan scaffold of each group was implanted into the SD-Rat full-thickness skin defect model with the same diameter of 1.5 cm. Then the regeneration tissues wereharvested and stained with hematoxylin and eosin and then evaluated microscopically. To verify the structures of hair follicles, immunohistochemical staining (IHC) was carried out.Results:In the experiment group, the HE staining of paraffin section shown some incompact hair follicle-like structures 10d post-surgery; with gradually increasing 21d post-surgery; and more mature hair follicle-like structures 35d post-surgery. The hair follicle-like structures post-surgery was positive for ALP by the immunohistochemical staining (IHC), which demonstrated the hair follicle specificity of these structures. The results illustrated that it could achieve the regeneration of epidermis and dermis with hair follicles at the same time in the experiment group. Correspondingly, in the other two control groups, it could achieve the regeneration of epidermis and dermis without the regeneration of hair follicles.Conclusion:1. The collagen/chitsan scaffold being incorporated with BMSCs could not achieve the regeneration of hair follicles.2. The collagen/chitsan scaffold being incorporated with HGF could not achieve the regeneration of hair follicles.3. The collagen/chitsan scaffold being incorporated with both BMSCs and HGF could achieve the regeneration of hair follicles.
