Background:Innate immunity is the body involved in anti-bacterial, anti-virus, anti-tumor first line of defense.Toll-like receptors (Toll like recepto rs, TLRs) is the key of innate immunity receptors, is also to connec t innate immunity and nonspecific immunity of the bridge, currently found ten members. Existing studies have found that the tumor ass ociated with TLRs have basal cell carcinoma, melanoma, bladder ca ncer, pancreatic cancer, breast cancer, head and neck cancer, etc, TL Rs play an important role in the process of the onset of these tumo rs. Activation of TLRs in tumor cells can induce the production of inflammatory factors and immune recation, while promoting inhibitio n of tumor cell proliferation and apoptosis, but also to promote tum or cell invasion and metastasis by metalloproteinase and quantity of the integrin protein. A study shows TLR9 associated with the occurr ence and metastasis of nasopharyngeal carcinoma in Chinese populati on, TLR9 CC-1486-t/C genotype can control sensitivity and tum origenesis of nasopharyngeal carcinoma (NPC) at the same time, als o can improve the expression of vascular endothelial growth factor i n nasopharyngeal carcinoma.Objective:To observe the distribution and expression of TLR9 (Toll like r eceptor-9, TLR9) in nasopharyngeal carcinoma tissue and adjacent no rmal tissue, and explore the role of TLR9 in the pathogenesis nasop haryngeal carcinoma pathogenesisMethods:Clinical data of 97 cases of pathologically diagnosed nasopharyngeal carcinoma were collected in this study.RT-PCR and Western blot analysis were used to detect the expression of TLR9 mRNA and protein levels between the nasopharyngeal carcinoma tissue and adjacent normal tissue. Immunohistochemical assay was employed to study the characteristic of distribution of TLR9 in the nasopharyngeal carcinoma tissue and adjacent normal tissue. Further, to evaluate the relationship of TLR9 between the clinical parameters and expression with nasopharyngeal carcinoma.Results:RT-PCR showed that the expression level of TLR9 was signific antly increased in nasopharyngeal carcinoma, compared to adjacent n ormal tissues with statistically significant (P<0.01).Western blot an alysis further supported the similar results in protein expression of T LR9 in the nasopharyngeal carcinoma tissues (P<0.01). The expressi on levels of TLR9 closely related of the clinical parameters of tumo r. Some cells showed very weak expression of TLR9 in adjacent no rmal tissue. Compared with adjacent normal tissue, nasopharyngeal ti ssue showed significant expression of TLR9, and the strong expressi on and distribution of some tumor cells and inflammatory cells in th e nucleus and cytoplasm in the nasopharyngeal carcinoma.Conclusion:The gene and protein expression levels of TLR9 in nasopharyng eal carcinoma were significantly increased closely associated with cli nical progression, indicating that TLR9 plays a role in the developed process of nasopharyngeal pathology;it may be as an important pr ognostic indicator in clinical judgment.
