| RATIONALES:Astragali Radix, owes a long use history. There always existed many researches against quality of Astragalus in three fields including Appearance, Biological traits and chemical point. According to the the aspect of chemical analysi, many studies mainly focus on the active ingredients of flavonoids and saponins in Astragalus. Those researches always just determined one or only few chemical components, furthermore, the influence factors to the quality just involve one aspects like different places or different planting mode,so the study always cannot reflect the quality comprehensive. The present study analyzed the differences of materials including different origins, planting mode, producing plants, grades and parts based on 10 chemical components.OBJECTIVE:By determining the contents of flavonoids and saponins, analyze the differences of the Astragalus,and furthermore, we can provid some suggestions according to the production and processing of applications.METHODS:Establish the sample preparation methods and detection method. Sleeted samples from different origins, planting mode, producing plants, grades and parts.Determine the contents of 10 kinds of index components,including 6 kinds of flavoniods concluding calycosin-7-O-β-D-glucoside(component 1), ononin(component 2), (6aR, 11aR)-9,10-dimethoxypterocarpan-3-O-D-glucoside(component 3), (3R)-2’-hydroxy-3’,4’-dimethoxyisoflavan-7-O-β-D-glucoside(component 4),calycosin(component 5),formononetin(component 6),and 4 kinds of saponins including astragolosides I (component 7), astragolosides II (component 8), astragolosidesⅢ component 9),astragolosidesⅣ(component 10), finally, appraised the quality of Astragalus with different origins, planting mode, producing plants, grades and parts.RESULTS:1.Preparation method:1.5g sample powder isaccurately weighed into theround bottom flask, add 60ml of methanol, reflux for 3 hours, filtered, recovery solvent to dry, anddissolved the residue in methanol, transfered to a 5ml volumetric flask, add methanol to the mark.Detection method:the mobile phase was acetonitrile (A)-water (B), the flow rate was 1 mL/min, ELSD parameters:N2 pressure was 23psi, atomization temperature was 35℃, vaporization temperature was 50 ℃, the gain value is 5, the Venusil MP C18 column (250 mm* 4.6 mm,5 m), the UV detection wavelength was 230nm, the column temperature was 25 ℃, gradient elution:0-8min,20%A; 8-15min,30%A; 15-30min,43%A; 30-40min,60%A; 40-50min,1 00%A; 50-60min,100%A; 60-65min,20%A, volume of sample 20ul.2.Study on the quality with various origins of AstralusThe present study selected two kinds of Astraglusand determined the thecomponents’contents. It said that, the contents of component 1, component 2, component 3, component 4, component 6, component 7, component 9 in the former were higher than in the later.,but the contents of componentcomponent 5, component 8, component 10 in the later were higher than in the former.3. Study on the quality with various planting mode of AstragalusThe study selected two kinds of pattern of Astragalus:cultivated and uncultivated,and determined the components’contents, it said that, the contents of component 1, component 3, component 4, component 5 and 4 kinds of saponins including component?, component 8, component 9, component 10 in the wild MongoliaAstragalus were higher than in the cultivated,but the components of component2, component 6 were the contrary. The contents of component 1, component 2, component 3, component 4, component 5, component 6 and 4 kinds of saponins including component 7 in the wild A.membranaceus (Fisch.)Bge. were higher than in the cultivated,but the contents of component8, component9, componentlOwere the contrary.4. Study on the quality with various producing areas of AstragalusThe present study selected wild MongoliaAstragalus from shanxi,niemong,shanxi provinces, cultivated MongoliaAstragalus from shanxi,gansu,niemong, provinces, cultivated A.membranaceus (Fisch.)Bge. from shanxi,hebei,Shandong provinces, and determined the contents of the 10 components.The result indicated that,the quality of Astragali Radix with various producing areas had some differences among them.Differences in Shanxi, Shaanxi, Inner Mongolia was little; to the cultivated, flavonoids of Shanxi origin the highest, saponins of Gansu was the highest; the chemical components in Shanxi County was the highest.5. Study on the quality with various grades of AstragalusThe present study determined the flavonoids and saponins’contents from yingxian Aatragalus with different grades. It said that, the ranking of the contents of componentl, component, component4,components, component6 in different grades of yingxian Astragali Radix were existed differences,the ranking of the contents of component3was the same withcomponentl.The sum contents of the 6 flavonoids from high to low class was fifth>second>third>fourth> first. To the saponins, the ranking of astragolosides I, astragolosides Ⅱ,astragolosidesⅢin different grades of yingxian Astragali Radix existed differences,the ranking of astragolosidesIV was the same with astragolosides Ⅱ. The sum contents of the 4 saponins from high to low grades was fifth> fourth> third>second> first.6. Study on the quality with various parts of AstragalusThestudy determined the flavonoids and saponins’contents in Astraglus including stem, basal part of stem, head part of root, main root, lateral root, fibrous root. It said that, the stem part had the highest contents about flavonoids and saponins, and the basal part of stem had the lowest contents about total flavonoids and saponins. To the different part, main root,lateral root and fibrous root, the main root had the lowest contents, the fibrous had the highest, but to the flavonoids, the fibrous had the lowest contents,the main root was the highest.CONCLUSION:1.The method is reliable, stable, and can be used for quality control of Radix Astragali.2.The quality of MongoliaAstragalus are better than A.membranaceus (Fisch.)Bge.And the result conformed the reality of the mainstream varieties of MongoliaAstragalus.3. The quality of wild Astragali Radix are better than the cultivated.4. In appropriate production area,specific ecological environment directly affects the chemical composition5.The relationship betweenflavonoids and Astragalus level is not strong, but the content of saponins are basically increased with the level of decreasing.6.The active ingredient content of the leaves are even higher than the roots, so the leaves have great prospects of development and application value. |
