Diabetes Mellitus was a syndrome characterized by hyperglycemia which led to glucose and lipid metabolic disorder. Patients suffered from this lifelong disease because of its characteristics of relapse. This mixture was a blend of the sea buckthorn leaves, mulberry leaves and whole gynura divaricata, the crushed mixture is crude drug. It obtained the three extract of mixture by different solvent (water, ethanol, methanol). Then investigated curative efficacy of four samples by Diabetes Mellitus model test, and screen out the effective samples. Afterwards, counted the proportion of each kind of blood cell and observed morphological changes of cells. HPLC was used to determine the constituents that have hypoglycemic effects of mulberry leaves, sea buckthorn leaves and gynura divaricata. It was provided a scientific standard for the mixture in its further research and manufacture. In experiment, the diabetic model was established with alloxan and high calorie diet. After experiment, checked the changes of physiological indexes of model of SD rats, and determined whether each sample can resist the damage. It included weight changes in experiment, fasting glucose and glucose tolerance, the content of blood lipids and insulin C-Peptide, the proportion of glycated hemoglobins. The experiment on the sixth, twenty-eighth and thirty-second day, drew blood by pricking skin of tail then processed blood smear. The microslides were satained by Wright-Giemsa then photographed, each kind of cells was observed identified and counted. Corosolic acid, Chlorogenic acid and Astragaloside have hypoglycemic effect, in which identitied from three plants at pretest. HPLC was developed for the determination of those components, and content were measured. The experiment suggested that aqueous extract could alleviate weight loss. The fasting blood glucose was declined by crude drug and aqueous extract. All of samples repaired impaired glucose tolerance, and declined glucose area under curve (GAUC). In aspect of blood-lipid regulation, aqueous extract and methanol extract declind cholesterol content and triglyceride content of SD rats. Ethanol extract improved index of triglyceride and high-density lipoprotein. In brief, three extracts had a function of regulating blood lipid level. In addition, aqueous extract and ethanol extract improved hyperinsulinemia and insulin resistance. Compared with model group, other groups did not significantly improve about glycosylated hemoglobin, in which implied that all drugs need a longer period to work. From observation of blood cells, it indicated that crude drug aqueous extract and ethanol extract improved the rise of proportion of mononuclear leucocyte. Aqueous extract and methanol extract improved the neutrophil hypersegmentation and lymphocyte chromatin condensation.The content of chlorogenic acid in sea buckthorn leaves, mulberry leaves and Gynura divaricata was respectively 8.753mg/g, 1.811mg/g and 5.864mg/g, content of corosolic acid was 0.550mg/g, 0.198mg/g and 0.012mg/g. The content of astragaloside in mulberry leaves was 21.23μg/g. In model experiment, it suggested that aqueous extract and ethanol extract regulated blood glucose and blood lipid effectively, they all improved insulin resistance and inflammatory state, in which due to hyperglycemia. The effectiveness of aqueous extract and ethanol extract were better than others. Aqueous extract and ethanol extract were suited to exploit as a kind of hypoglycemic heath food. By HPLC determining astragaloside and corosolic acid content were accurate, convenient and reliable, which were suited to be the standards of quality control.
