BackgroundAcute myocardial infarction(AMI) is the most severe clinical phenotype in patients with coronary heart disease(CHD). Coronary artery blood flow, which occurs on the basis of coronary heart disease, is rapidly reduced or interrupted, which leads to the corresponding myocardial ischemia and hypoxia. High fatality rate, pose a serious threat to human life and health AMI and CHD are a group of diseases affected by various environmental factors and genetic factors, a large number of data show that genetic factors play an important role in the pathogenesis of this group of diseases. However, as coronary atherosclerotic heart disease,it is still not clear whether genetic factors are involved in the process of the evolution from stable angina pectoris(Stable angina pectoris SA) to acute myocardial infarction. we analyzed the difference of the peripheral blood RNA expression profiling of AMI and SA patients and study which gene may be involved in the process of transformation from SA to AMI in our study, so as to we can find out the genetic marker of acute myocardial infarction, and to lay the foundation for the diagnosis and treatment of acute myocardial infarction. ObjectiveIn order to investigate genetic change in the incidence of acute myocardial infarction, to find the gene markers warning the occurrence of acute myocardial infarction, and to screen the differentially expressed genes in patients with coronary atherosclerotic heart disease from stable angina pectoris to acute myocardial infarction, We applied c DNA microarray to detect peripheral blood gene expression profiles of the patients with acute myocardial infarction and stable coronary heart disease.MethodA total of 30 patients with acute myocardial infarction and stable coronary heart disease were selected, who were admitted to the China Japan Union Hospital of Jilin University between March and November 2014-2015. There are 15 patients with stable angina and 15 patients with acute myocardial infarction. MRNA in peripheral blood white blood cells were extracted and reverse transcribed into c DNA. Detection of protein in peripheral blood leukocytes by blot western. Three patients in each groups were randomly selected for gene chip detection, and the other samples were used to verify the results of gene chip. TTC7 B and PRMT5 genes were randomly selected from the high expression of gene chip, and the GTPBP4 was selected as the no difference group, and the CPNE3 was selected as the low expression group. We applied Real time fluorescent quantitative PCR to detect the selected genes and analysis of the differential expression of these genes in different disease groups. TTC7 B gene was detected by blot western, and the protein expression level of TTC7 B gene in different groups was analyzed. ResultGene chip detection results showed that the two groups of patients with a total of 16 differentially expressed genes, 2 low expression genes, 14 highly expressed genes(p-value < 0.05, |log FC| > 1). q PCR shows, TTC7 B gene in acute myocardial infarction group expression is higher than in stable coronary artery disease group, and the difference is significant(P < 0.05); Prmt5 gene in acute myocardial infarction group the expression is higher than that of stable angina pectoris(SA) group, and the difference is significant(P < 0.05); CPNE3 gene in the acute myocardial infarction group expression is lower than that in stable coronary heart disease group, and the difference was significant(P < 0.05); GPTBP4 gene in myocardial infarction group expression and stable angina pectoris group exist no difference(P > 0.05); Western blot results shows: The expression of TTC7 B in myocardial infarction group is higher than in patients with coronary heart disease. ConclusionGene chip result is true and reliable; The peripheral blood RNA expression levels in patients with acute myocardial infarction and stable angina pectoris are different, There are 16 differentially expressed genes in the two groups, in which there are 2 low expression genes and 14 highly expressed genes; TTC7 B, Prmt5, cpne3 may be genetic markers for diagnosis of acute myocardial infarction.
