Given that a large propotion of genes encode non-coding RNAs, more and more people focus on the regulatory roles of them in recent years. MicroRNA (miRNA) is a kind of single-strand noncoding RNAs,which is 18 to 24 nucleotides in length and encoding by endogenous gene. They are involved in post-transcriptional regulation of gene expression. miR-17-5p,20a belong to miR-17 family and miR-17-92 cluster. miR-17 family has been reported highly expressed in tumor cells, implying its signaficance in tumorigenesis and aging. In this project, we inhibited miR-17 family expression through decoy technology in breast cancer to investigate effects on tumor cell proliferation, migration and invasion,MiR-17 family includes eight micrornas. There is a conserved sequence from the second to the eighth nucleotide at the 5’end of the mature miRNA, referred to as the seed sequence. We reduced miR-17 family microRNA expression in breast cancer cells by decoy technology through lentivirus system. After the infection of the decoy miR-17 vector, we investgiate whether decoy 17 can inhibit tumor cell proliferation by recording tumor cell proliferation and drawing cell growth curve. And we defined whether decoy 17 effects on tumor cell proliferation and migration through the wound-healing and transwell experiments. Furthermore,we validated the experimental results in mice, and exploring the mechanism of the miRNA function through downstream target proteins.
