Objective: To investigate the expression of long noncoding RNA of bladder cancer associated transcript 1(non-protein coding)(blacat1)in different WHO graded glioma specimens and effects of long noncoding RNA blacat1 on basic biological function of such as glioma proliferation,migration and invasion.Methods: 85 cases of gliomas obtained by surgery from February 2013 to February 2017 were classified into low-grade glioma(I?II)group(25 cases)according to WHO classification(I?IV),high-grade glioma(III?IV)group(45 cases),and 15 cases of brain tissue samples from trauma patients were used as normal group.Real-time quantitative Polymerase Chain Reaction(qRT-PCR)was used to detect the expression of long non-coding RNA BLACAT1 in glioma samples and normal tissue samples,and to analyze the correlation between long non-coding RNA BLACAT1 and glioma WHO classification.The transfected glioma cell lines LN382 and U87 MG were divided into silence group(transfected long non-coding RNA BLACAT1 siRNA)and NC group(transfected negative siRNA)by small interfering RNA transient transfection technique.RTCA proliferation test,scratch test,Transwell migration /invasion test were used to detect the changes of glioma cell proliferation,invasion and migration in silence group and NC group.Results: The higher the WHO classification of glioma samples,the higher the expression of long noncoding RNA BLACAT1(p<0.01).In the RTCA proliferation experiment,the proliferation rate of the silence group was significantly slower than that of the NC group after 120 hours(p<0.01).In the cell scratch test,the scratch healing rate of the silence group measured at 12 hours and 24 hours was significantly slower than that of the NC group(p < 0.01).In the Transwell invasion/migration experiment,we measured that the number of basement membranes passing through the compartment at 24 hours in the silence group was significantly less than that in the NC group(p<0.01).Conclusion: The higher the grade of gliomas,the higher the expression of long noncoding RNA BLACAT1.The long noncoding RNA BLACAT1 promotes the basic biological functions such as proliferation,migration and invasion of glioma.
