Influence Of Rhizoma Nelumbinis On P-JAK2, P-STAT3 And VEGF Of Renal Tissue In The Diabetic Nephropathy Rats

Objective:To investigate the influence of rhizoma nelumbinis on p-JAK2, p-STAT3 and VEGF of renal tissue in the diabetic nephropathy rats.Methods:60 SD male rats were divided randomly into control group(group C), diabetic nephropathy group(group DN), rhizoma nelumbinis Low-dose treatment group(group RL, 1.5g·kg-1·d-1), rhizoma nelumbinis Dose treatment group(group RD, 3.0g·kg-1·d-1), rhizoma nelumbinis High-dose treatment group(group RH, 6.0g·kg-1·d-1), Losartan treatment group(group L, 30mg·kg-1·d-1). The rats were being intraperitoneal injected with STZ (1%,45mg/kg) to establish diabetic nephropathy models.The standard of building diabetic nephropathy rats was that random blood glucose test> 16.7 mmol/L, and 24h urinary albumin> 30mg · kg-1· d-1 after 3 weeks. All rats received daily gavages before dissected, test period 12 weeks. After 12 weeks, blood glucose, body weight, the 24h urinary protein, serum creatinine (Scr), urea nitrogen(BUN) were measured. The renal pathological changes were detected by light microscope and electron microscopy. The expression of p-JAK2, p-STAT3 and VEGF was measured by immunohistoche mistry staining sections.Results:After establishing mode for 12 weeks, compared with group C, group DN had significant elevated 24-hour urinary protein, Blood glucose, Scr and BUN (P<0.05), and immunohistochemistry staining displayed that the expression of P-JAK2, p-STAT3, VEGF remarkably increased(P<0.05). The difference of 24-hour urinary protein, Blood glucose, Scr, BUN, the expression of p-JAK2, p-STAT3, VEGF between group RL and DN were not statistically significant. Compared with group DN, group RD, group RH had significantly lower 24 urinary and the expression of p-JAK2, p-STAT3 and VEGF were lower than those in group DN(P<0.05). The effect of groupRD and group RH were similar to each other(P> 0.05).Conclusion:rhizoma nelumbinis might in part through regulating the expression of P-JAK2, p-STAT3 and VEGF play a protective role in the kidney of diabetic nephropathy rats.
