Objective: In order to investigate that weather the protective effects of GIK solution combined with glutamine between respectively alone use have differences,We established endotoxemic intestinal injury of rat models by intraperitoneal injection lipopolysaccharide, and to explore its possible mechanisms.Methods: 90 SD rats were randomly divided into five groups: Normal saline group(NS,n=10),lipopolysaccharide group(LPS);Glucose- GIK-potassium solution group(GIK);glutamine group(Gln);GIK+Gln group(n=20 each).Endotoxemia was induced by lipopolysaccharide(LPS) injection, the glutamine(2g/d) was immediately administered with oral after LPS injection;GIKsolution(G 200g/l insulin :60 IU/l,KCL:60mmol/l) was intravenous infusion(4 ml/kg/h)after 2 hours of LPS injection.Plasma and small intestine tissues were collcted after 3days.The TNF-α and DAO levels of plasma were detected by ELISA,The plasma D-lac were measured by biochemical methods.The intestine tissues were made into paraffin section and it’s pathological changes were observed by light microscope,meanwhile evaluated the injury degree by Chiu’s 6 grade score.Results: 1.Except The rats in NS group, after intraperitoneal injection of LPS 0.5h-1h,all rats appear fatigue, jitter,reduced activity, after 24 h,GIK group,Gln group and GIK+Gln group were improved,meanwhile the LPS group appear to eat less,less activity,drowsiness,abdominal distension, diarrhea, etc.2.The TNF-α level of LPS group,,GIK group,Gln group and GIK+Gln group were higher than NS group(p<0.05),and the LPS group was the highest.3. Compared with normal contro I group,The plasma D-lac and DAO were significantly increased in LPS group(p<0.05). The levels of D-lac and DAO in GIK group,Gln group and GIK+Gln group were lower than those in LPS group(p<0.05).There was no difference between GIK group and Gln group(p>0.05).In contrast,the D-lac and DAO activity in GIK+Gln group were significantly reduced(p<0.05).4. Compared with normal contro I group,the Chiu’s grade were significantly lower in others groups.But there was no significantly difference among GIK group,Gln group and GIK+Gln group(p>0.05).Conclusion: 1.Endotoxemia induced by LPS in rats can lead to intestinal injury,resulting in damage to intestinal cells and mucosal barrier dysfunction. 2.There were protective effects in rats of endotoxemic intestinal injury models by intraperitoneal injection LPS treatmented with GIK.But there was no significantly difference among GIK group and Gln group.When both of GIK and Gln were used,the effects were significantly better...
