The Danxiao Tea Effects On IGT Rats Expression Of TNF-α IL-6 And GLUT In The Liver And Skeletal Muscle

Objectives:Use Danxiao Tea to cure IGT rats,observe the changes of TNF-a, IL-6 and liver GLUT-2 and skeletal muscle GLUT-4 mRNA expression,then explore the mechanism of Danxiao Tea treatment of IGT rats,in order to provide new methods and experimental basis for prevention and control of the IGT.Methods:1.male SD rats,using high sugar.and high fat diet and injecting 2% STZ(20mg/kg)copy the model of IGT,when blood glucose<5.6mmol/L,and in 7.8-11.1 mmol/L 2h after the OGTT,the IGT model is succeed.2.Model rats are randomly divided into 4 groups:the model group of IGT,the treatment group of Metformin Tablets,the treatment group of Danxiao Tea and the group of sports therapy;and set a normal group at the same time.3.Use the drugs to cure rats:feed the treatment group of Metformin Tablets with Metformin aqueous solution;feed the treatment group of Danxiao Tea with the aqueous solution of Danxiao Tea;train the group of sports therapy with the modified Ploug.give the normal group and model group just distilled water.4.Observe the general situation in rats,after 8weeks treatment,check the Glucose、HbAlc、FINS、TC、TG、HDL-C、LDL-C、TNF-α、 IL-6.Use the RT-PCR to detect GLUT-2 mRNA in liver,GLUT-4 mRNA in skeleton muscle.And take the liver pathological examination.Result:Danxiao Tea can reduce the expression of TNF-a、IL-6 and improve the expression of GLUT-2,GLUT-4 mRNA in IGT rats,also can reduce the lipid metabolism in the rats so as to alleviate the insulin resistance,lower the glucose and HbAlc,thus cure the IGT condition.Conclusions:In this experiment,we copy the IGT models and use the Danxiao Tea to cure the IGT rats,observe the changes in TNF-a,IL-6 and GLUT,then get some conclusions below:1.the TNF-a and IL-6 has a significant increase in IGT rats,remind that the abnormal expression of two inflammatory cytokines hint in IGT lesions plays a certain role in the process.2.the GLUT expression in liver and skeletal muscle decreased significantly in the IGT rats than the normal group,remind that the abnormal expression of GLUT in IGT pathological changes play a relevant role in the process.3.After the intervention of Danxiao Tea,the TNF-a,IL-6 and blood lipid decreased obviously,the expression of GLUT rise obviously,blood glucose and HbAlc decreased obviously,and prompt the TNF-a,IL-6 and GLUT lesion degree of correlation with IGT.4.Danxiao Tea,the effects of the treatment of IGT rats may be by reducing the TNF-a and secretion of IL-6,reduce blood fat,and raised the expression of GLUT,so as to achieve the effect of lowering blood sugar.
