Abstract/Summary:
ObjectiveTo discuss mechanisms of Tongluo Decoction(TLP) against D-GaIN-induced acute liver failure(ALF)rats with heat toxin and by Observation of ALF biochemical rat rat model, changes in survival time, pathology, TLR4, NF-K B, HMGBI, Caspase-3, and other indicators.MethodsHeal the clean grade SD rats, were randomly divided into four groups, that was normal group(B), model group, SNMC group and TLP group injected intraperitoneally with D-GaIN to induce ALF rat model.Model group, SNMC group and TLP group were injected intraperitoneally with D-GaIN 1.49/kg.SNMC group and CLP group were respectively treated with 1 5.6mg/kg/d and 29.79/kg/d. The survival rate of rats at hour 96 were mainly observed.serum ALT, AST, TBiL, ALB and CHE levels were measured via automatic biochemical analysis; plasma PT was measured via automatic coagulation analysi s; express ion levels of TLR4、NF-κB、HMGBI and caspase -3 mRNA in the 1 ivert i S sue were detected Via RT-PCR fluorescence method.Result1. Survival rate:after 96h there are 33.3% who survived in model group.46.7% in SNMC group and 53.3% in TLP group. There was no significant difference(P>0.05)in comparing the survival and death rate among the three groups.At 96 hours, mean survival times of the three groups were 63.5,71.8 and 83.8 hours respect ively; cumulative survival rate of rats in CLP group was higher than that of rats in model group(P<0.05).However, there was no significant difference between SNMC group and model group or CLP group(P>0.05).2. The levels of ALT, AST:Compared with B group, the levels of ALT, AST in M group,P group and TLP group were increaseds significantly;Compared with model group,the levels of ALT, AST in P group and TLP group were decreased significantly. Compared with SNMC group,the levels of ALT, AST in TLP group were decreased significantly. There were significant differences(P<0.05).3. The levels of TBIL:Compared with B group, the levels of TBIL in M group,P group and TLP group were increaseds significantly;Compared with model group,the levels of TBIL in P group and TLP group were decreased significantly. There were significant differences(P<0.05).While, there was no significant difference between SNMC group and model TLP group(P>0.05).4. The levels of ALB、CHE、PT:Compared with normal group,the levels of ALB、CHE in model group,SNMC group and TLP group were decreaseds significantlyin, while the PT inthese groups were increaseds significantly.There were significant differences(P<0.05).Compared with model group,the levels of ALB, CHE、PT in TLP group were ameliorated significantly. There were significant differences(P<0.05).5.The relative content of TLR4 mRNA expression in rats liver tissue:Compared with normal group,the relative content of TLR4 mRNA in model group were increaseds significantly.There were significant differences(P<0.05).Compared with model group, the relative content of TLR4 mRNA in both SNMC group and TLP group were decreaseds significantly.And, and the decrease in TLP group was greater.There were significant differences(P<0.05).6. The relative content of NF-κ B mRNA expression in rats liver tissue:Compared with normal group,the relative content of NF-κB mRNA in model group were increaseds significantly.There were significant differences(P<0.05).Compared with model group, the relative content of NF-κB mRNA in both SNMC group and TLP group were decreaseds significantlyAnd, and the decrease in TLP group was greater.There were significant differences(P<0.05).7. The relative content of HMGBI mRNA expression in rats liver tissue: Compared with normal group,the relative content of HMGBI mRNA in model group were increaseds significantly.There were significant differences(P<0.05).Compared with model group, the relative content of HMGBI mRNA in both SNMC group and TLP group were decreaseds significantly.And, and the decrease in TLP group was greater.There were significant differences(P<0.05).8. The relative content of Casepase-3 mRNA expression in rats liver tissue: Compared with normal group,the relative content of Casepase-3 mRNA in model Group were group were increased sigificantly(P<0.05).Compared with model group, the relative content of Casepase-3 mRNA in both SNMC group and TLP group were decreaseds significantly.And, and the decrease in TLP group was greater.There were significant differences(P<0.05).ConclusionTongluo Decoction cound increase the survival time and the survival rate of rats significantly with acute liver failure, cound improve their liver function, and promote the repair of coagulation and promote the regeneration of liver cells. In the other side,Tongluo Decoction cound decrease TLR4 and HMGBI’s expression of liver tissue;block the signaling pathway of TLR4/NF-κB; reduce the expression of pro-inflammatory cytokines, decrease the expression of Caspase-3mRNA, apoptosis of liver cells promote the regeneration of liver cells.
